Objective:In this study,we investigated the expression of YEATS4(YEATS domain containing protein 4)in pancreatic cancer tissues,and analyzed the relationship between YEATS4 protein expression and clinicopathological features and prognosis of pancreatic cancer patients,and explored its clinical characters in the diagnosis and treatment of pancreatic cancer.Methods:This study collected clinical data and paraffin specimens from 80 patients with pancreatic ductal carcinoma(PDAC)who underwent surgical resection and pathological diagnosis from March 2011 to August 2017 in Jiangsu University Hospital.Immunohistochemistry technique was used to detecte the expression of YEATS4 protein in all samples.Eight pairs of samples were randomly selected from all the samples to detect the expression of YEATS4 protein in pancreatic cancer and its matched normal tissues by Western Blotting technique to further verify the results of immunohistochemistry.SPSS19.0 statistical software was used to analyze the correlation between the expression of YEATS4 in pancreatic cancer tissues and the clinicopathological parameters of pancreatic cancer and the prognosis of patients with pancreatic cancer.Result:The study found that: 1.Compared with normal pancreatic tissue,the positive expression of YEATS4 in pancreatic cancer tissues was significantly increased,and the difference was statistically significant(P<0.05).2.The expression of YEATS4 protein in pancreatic cancer was significantly higher than that in adjacent normal tissues.3.The positive expression rate of YEATS4 was associated with tumor TMN stage(P<0.001),neurotransmission(P=0.006),lymph node metastasis(P<0.001),and vascular metastasis(P=0.042)in patients with pancreatic cancer.4.Survival analysis showed that the YEATS4 protein negative expression group had longer survival than the positive expression group(P<0.05).5.Univariate and multivariate regression analysis further confirmed that YEATS4 protein expression can be used as an independent risk factor for prognosis in patients with pancreatic cancer.Conclusion:The expression of YEATS4 in pancreatic cancer is significantly increased,which may be involved in the progression of pancreatic cancer.The poor prognosis of patients with pancreatic cancer is associated with an abnormal increase in YEATS4 expression.The expression of YEATS4 is expected to be a new potential tumor marker in the diagnosis of pancreatic cancer and potential molecular target for drug therapy.
