| Objective:The essay was aimed to investigate the expression of Nup88 gene in prostate cancer tissues and the biological behavior of DU145 prostate cancer cells in vitro and in vivo,and to determine whether this gene can be used as a potential target for the diagnosis and treatment of prostate cancer and offers a new direction for castration resistance prostate cancer.Methods:The experiment was carried out in three steps.First,The expressions of Nup88 gene in prostate cancer DU145 cells and normal prostate epithelial RWPE-1 cells were detected by qRT-PCR and Western blot.The specimens of 18 cases of castration-resistant prostate cancer tissues and adjacent tissues were detected by immunohistochemistry to evaluate relative expression level of Nup88.Secondly,LV-NUP88 shRNA-GFP vector was transfected into DU145 cells to down express of Nup88 gene.The expression of Nup88 protein was detected by Western blot.cell proliferation was detected by CCK-8.cell migration and invasion ability were detected by scratch test and Transwell test,and cell cycle were detected by flow cytometry.Finally,the experimental group(LV-NUP88 shRNA-GFP),the negative control group(LV-GFP),and the blank control group were planted into three nude mice groups randomly.The transplanted tumor was collected on the 40 th day and weighed and analyzed.Results:In the first part,qRT-PCR and Western blot showed that the expressions of Nup88 mRNA and protein in DU145 cells were significantly higher than that in normal prostate epithelial RWPE-1 cells.The immunohistochemical results of prostate cancer tissues and adjacent tissues showed that the expression of Nup88 protein in prostate cancer tissues was significantly higher than that in adjacent tissues.In the second part,the down-expression of Nup88 gene in DU145 was obvious in the experimental group.The results of CCK-8,scratch test,Transwell experiment and flow cytometry showed that the cell proliferation,migration were weakened.The cells are mainly arrested in the G1 phase.In the third part,the DU145 cells treated in three ways were successfully implanted in nude mice.The tumor weight analysis showed that the weight of the transplanted tumor was in the blank control group for40 days.There was no significant difference between the control group and the negativecontrol group.The weight of the transplanted tumor in the experimental group was smaller than that of the other two groups(p<0.05).Conclusions:The expression level of Nup88 gene in the prostate cancer tissue was higher than that in the adjacent tissue,which may promote the growth of prostate cancer cells.The expression of targeted silencing Nup88 gene inhibited the biological behavior of DU145 cells of prostate cancer.The Nup88 gene can serve as a potential target for targeted therapy of castration-resistant prostate cancer genes. |
