Study On The Relationship Between The Expression Of ZIP4 And Postoperative Survival In Patients With Hepatocellular Carcinoma

Background Liver cancer is one of the most common malignant tumors in China.Hepatocellular carcinoma(HCC)accounts for more than 50% of the world's primary liver cancer,and hepatocellular carcinoma(HCC)accounts for more than90% of HCC.At present,surgical treatment and liver transplantation are the main methods for treatment of hepatocellular carcinoma(HCC).Due to the lack of early clinical manifestations and the absence of specific molecular markers in patients with hepatocellular carcinoma(HCC),Therefore,most of the patients with hepatocellular carcinoma are in the middle and late stages of diagnosis and have lost the chance of surgery.Therefore,it is very important to find a new molecular marker and a new therapy for the diagnosis and treatment of hepatocellular carcinoma.Objective To investigate the role of ZIP4 in hepatocellular carcinoma and its relationship with the prognosis of patients with hepatocellular carcinoma.Methods The levels of ZIP4 mRNA in hepatocellular carcinoma cell line BEL-7402,HepG2 and normal liver cell line Changliver were detected by RT-qPCR,and the expression level of ZIP4 protein in cell line was detected by WB.Collection of pathology in patients with hepatocellular carcinoma from 2012 to 2013 The expression of ZIP4 protein in cancer tissues and paracancerous tissues was detected by immunohistochemical method in 62cases(including tumor tissues and paracancerous tissues).The relationship between the expression of ZIP4 protein and the clinicopathological features of HCC patients was analyzed.Kaplan Meier survival curve method was used to analyze the relationship between the expression of ZIP4 protein and the survival time of the patients.Results The expression of ZIP4 mRNA in two hepatocellular carcinoma cell lines and one normal liver cell line was detected by RT-qPCR.The results showed that ZIP4 mRNA was expressed in BEL-7402 cell line and HepG2 cell line.Theexpression of ZIP4 protein in the cell line was significantly higher than that in the normal liver tissue cell line Changliver.The expression of ZIP4 protein was detected by WB protein level,and GAPDH was used as the internal reference.The results showed that the expression of ZIP4 protein in hepatocellular carcinoma cell line BEL-7402 and HepG2 was significantly higher than that in normal human hepatocellular carcinoma cell line Changliver(P < 0.05).)The results of immunohistochemistry showed that the expression of ZIP4 protein was significantly higher in HCC than in paracancerous tissues,and the high expression of ZIP4 was negatively correlated with the median survival time of the patients.Conclusion The expression of ZIP4 in liver cancer cell lines and hepatocellular carcinoma tissues was significantly increased,which can be used as a marker of poor prognosis of liver cancer.
