Objective:To investigate the expression of platelet-derived growth factor BB(PDGF-BB)/receptor beta(PDGFR-?)and related signaling pathway factors in Oral Squamous Cell Carcinoma,we analyze the role of PDGF-BB/PDGFR-? and related signaling pathways factors in Oral Squamous Cell Carcinoma and its relationship with the pathological features of Oral Squamous Cell Carcinoma,in order to provide reference for the prevention and treatment of Oral Squamous Cell Carcinoma.Methods:(1)Case collection:84 cases of Oral Squamous Cell Carcinoma paraffin-embedded specimens were selected and confirmed by pathological diagnosis,the paracancerous normal tissues were used as control.(2)Experimental grouping:Pathological sectioning and HE staining were performed on the included Oral Squamous Cell Carcinoma tissues,and the cases were confirmed by pathological diagnosis.Based on this,the cases were divided into two groups:?Paracancerous normal tissue;?Oral Squamous Cell Carcinoma group;The tissue classification was determined according to the degree of cell differentiation,the cases were divided into three groups:? Highly differentiated group;? Moderately differentiated group;? Poorly differentiated group;lymph node metastasis of Oral Squamous Cell Carcinoma was determined according to the pathological examination results of lymph node disease,the cases were divided into two groups:?No lymph node metastasis group;?Cervical lymph node metastasis group.(3)Experimental methods:EnVision immunohistochemistry was used to detect the expression of a-SMA,PDGF-BB,PDGFR-P,TAB3 and NF-?B in OSCC tissue samples.(4)Results interpretation:according to the recent literature reports by Deepa Rajesh Mane and other scholars on the analysis of immunohistochemistry results[1],the results of results of immunohistochemical images were analyzed by using Image J 1.50i professional image software,it shows:Negative(-);Low positive(+);Positive(++);High positive(+++).(5)Data processing:Statistical analysis was performed on the data using SPSS 22.0 software.The data adoption rate and composition ratio were used.The grade data was analyzed by non-parametric Wilcoxon rank sum test.The difference between groups was analyzed by Kruskal-Wallis test with non-parametric test(H test),the correlation analysis was performed using Spearman rank correlation analysis,and the difference was statistically significant at P<0.05.Results:(1)HE staining showed that OSCC tissues was divided into three different degrees of differentiation:high-differentiation was similar to normal squamous epithelium,keratinization was obvious,and mitotic figures were few;moderate differentiation had unique nuclear polymorphism and mitotic figures,angle is not common;poorly differentiated people are mainly immature cells,with a large number of normal or abnormal mitotic figures,and very little keratinization.(2)The results of immunohistochemistry showed that a-SMA,PDGF-BB,PDGFR-?,TAB3,NF-?B and p-NF-?B were overexpressed in OSCC tissues,and all of them were positive expression,the expression of them in OSCC tissues was significantly stronger than that in paracancerous normal tissues,and the difference was statistically significant(P<0.001).a-SMA and PDGFR-? were mainly expressed in cancer-associated fibroblasts in the stroma,while in normal fibroblasts were at a low expression level;PDGF-BB was expressed in tumor cells and stromal cells,mainly expressed in tumor cells;TAB3,NF-?B,p-NF-?B were expressed in tumor cells;by Spearmann rank correlation analysis:a-SMA There were differences in the expression of OSCC tissues with different degrees of differentiation,and were correlated with the degree of differentiation(P<0.05).The expression of PDGF-BB,PDGFR-?,TAB3,NF-?B and p-NF-?B in OSCC tissues was associated with lymph node metastasis(P<0.05).(3)Further analysis of the results of immunohistochemistry by statistical software SPSS 22.0 showed that the expression of PDGF-BB was positively correlated with the expression of a-SMA,PDGFR-?,TAB3,NF-?B,p-NF-KB in OSCC tissues(P<0.05).Conclusions:Overexpression of PDGF-BB/PDGFR-? in Oral Squamous Cell Carcinoma may be an important factor in inducing fibroblasts into cancer-associated fibroblasts,it may be one of the important factors affecting cervical lymph node metastasis in Oral Squamous Cell Carcinoma;the molecular mechanism of PDGF-BB overexpression in Oral Squamous Cell Carcinoma may be related to the activation of NF-?B signaling pathway.
