Objectives:Polymorphisms of methylenetetrahydrofolate reductase(MTHFR)gene is one of the genetic factors leading to ESA.Relevant literature reports show that polymorphism of MTHFR gene is closely related to ESA in many regions at home and abroad,but the study on polymorphism of MTHFR gene and ESA in Yunnan has not been reported.Therefore,this study focused on the relationship between polymorphisms of MTHFR 677 C>T and 1298 A>C genes and ESA in Yunnan Province.Methods:200 persons with ESA were chose,Blood genomic DNA kit was used to extract and purify venous blood DNA.The polymorphisms of MTHFR 677 C>T and 1298 A>C genes were detected by double fluorescence quantitative PCR in Yunnan patients with ESA.The relationship between MTHFR 677 C>T and 1298 A>C genotypes and ESA was analyzed with the 193 healthy control group.Results:The frequency of MTHFR 677 C/C and 677 C/T genotypes was significantly different between the patients with ESA and the healthy control in Yunnan(P=0.00,P<0.05),while the frequency of MTHFR 677 T/T genotypes was not significantly different between two groups(P=0.416,P>0.05).The frequency of MTHFR 677 C/T genotypes in patients with ESA(47.5%)was significantly higher than that in healthy people(29.0%).There was no significant difference in polymorphism of MTHFR 1298 A>C gene between two groups in Yunnan(P>0.05).Conclusions:This study suggests that polymorphism of MTHFR 677 C>T gene is one of the main factors causing ESA in Yunnan.Because the number of MTHFR 1298 A>C samples collected is too small and the frequency of MTHFR 1298 C/C genotype distribution is too low,the relationship between polymorphism of MTHFR 1298 A>T gene and ESA in Yunnan needs further study.Objectives:Polymorphisms of methylenetetrahydrofolate reductase(MTHFR)gene are main genetic cause of early spontaneous abortion.A number of studies have shown that MTHFR 677C>T Polymorphism is one of the main causes of ESA,However,there is still debate on whether MTHFR 1298 A>C Polymorphism could affect ESA.For the lack of extensive research in the Chinese population at present,the study aim to investigate the relationship between MTHFR 1298A>C polymorphism with ESA through a large number of data.Methods:Blood genomic DNA kit was used to extract and purify DNA from venous blood of 241 patients with ESA and 117 healthy people in Yunnan.MTHFR 1298 A>C Polymorphism was detected by double fluorescence quantitative Polymerase Chain Reaction(PCR).Considering the low frequency of MTHFR 1298 C/C genotype,which may affect the results of statistical analysis,we further search the articles that MTHFR 1298 A>C Polymorphism related to ESA in Chinese population in CNKI,WanFang,WeiPu databases to obtain more accurate results.Results:No statistical difference was detected in the frequency of MTHFR 1298 A>C polymorphism between the persons with ESA and healthy control group in Yunnan.Our data showed that the frequency of MTHFR 1298A/A genotype had the decreasing tendency among Chinese population from northern to southern,as well as eastern to western of China.The frequency of MTHFR 1298 A/C and 1298 C/C genotypes had the adverse tendency,which had the increasing tendency among Chinese population from northern to southern,as well as eastern to western of China.Frequency of MTHFR 1298 C/C genotype was significantly different between two groups in Chinese populations.The significant difference was also observed in the frequency of MTHFR 1298 C/C polymorphism between two groups from Central China and Southern China.Conclusions:The study revealed that MTHFR 1298 C/C genotype was one of the most important factors for ESA in Chinese populations,and the lower frequency of MTHFR 1298 C/C genotypes in Chinese populations and small sample size of different study,which might cause the dispute of the role of MTHFR 1298 C/C genotypes on ESA in Chinese populations.
