ObjectiveBy analysising the genetic diversity and population genetic structure,Trying to find some locus corresponding to their geographic locations,furthermore attempting to relate geographic differentiation to the primary crime scene deduction by Forensic verification.MethodsThe complete sequence of the mitochondrial genome of the Chrysomya megacephala was detemined by using PCR and the first generation sequencing method.From south to north,this experiment collected 64 sample data in total in 10 areas and 16 sampling points.Different geographical populations were clustered by PCoA analysis results.Mantel test reveales significant correlation genetic and geographical distance.Using molecular variation analysis method(AMOVA)analysis of population genetic differentiation between index Fst and Fst of significance test method.Fu sFs values are calculated by use of a neutral inspection,to infer population expansion history.Through the multiple sequence alignment statistical induction type is closely related to the geopolitical relations of haploid type.ResultsAnalysed from the average genetic distance,The fastest rate of evolution is ND3,ND4 gene,followed by CYTB gene,ATP8 is a very conservative and no variable sites were found.The NeiMeng population in High Latitude Areas gives a both negative value,showing that the population holds a expansive trend.Mantel test also revealed significant correlation genetic and geographical distance.Molecular variation analysis(AMOVA),Fst=0.23208(P<0.01),showed that 76.79% of the variance was from interpopulation,and 23.21% of the variance was from intrapopulation,indicating significant genetic differentiation between the 10 populations.PCoA analysis result and phylogenetic tree constructed using NJ and MX methods and network structure showed that different geographical populations were clustered into 3 subspecies divided by A,B,C.Through sequence alignment,two haplotypes produced by mutation and non mutation on the 4785 and 5804 sites of the A subspecies were clustered corresponding to their geographical distribution.The rRNA gene squence among Yunnan populations showed a slower evolutionary trends,which embodies the sites on 13533,14304,14305,14521,14849 is more stable while other populations is instable.The ND5 coding genes among Neimeng populations showed a higher Nucleotide diversity than others.ConclusionsThrough the analysis of biological genetic diversity,there are obvious differences among geographical populations,and the sequence differences between different geographical samples can reflect the geographical position to some extent.It can provide theoretical basis for forensic scientists to infer the first scene of murder through forensic insects rely on the two haplotypes produced by mutation and non mutation on the 4785 and 5804 sites of the A subspecies were clustered corresponding to their geographical distribution and the Haplotypes of Yunnan regional population among rRNA coding region.It provides a new way to improve a further study of forensic entomology and its inference in the site of death.
