Objective:Thrombspondin-1(TSP-1)is a multifunctional extracellular matrix glycoprotein expressed in various cells and participate in a variety of physiological activities,including inhibition of angiogenesis,involved in cell adhesion,migration,apoptosis.However,there is less research on the the role of induction of immune tolerance and graft versus host disease.We determine the expression of TSP-1 on the surface of dendritic cells(DCs)when acute graft-versus-host disease(GVHD)occurred in patients with malignant hematologic disease after allogeneic hematopoietic stem cell transplantation(allo-HSCT),and explore the relationship and significance between TSP-1 and acute graft-versus-host disease.Methods:take the patients who underwent allogeneic hematopoietic stem cell transplantation in clinic as the research object.We randomly draw peripheral blood before transplantation,with no aGVHD after transplantation,and with aGVHD just happened after transplantation but not aGVHD treatment.The mononuclear cells were isolated from peripheral blood of patients and be cultured to mature dendritic cells.The expression of TSP-1 on dendritic cell surface was detected by flow cytometry before transplantation,after transplantation with no aGVHD,after transplantation with aGVHD,compare the results to see whether the expression changes are significant.Results:The percentage of TSP-1 positive cells in the dendritic cells before transplantation was(47.92±9.24)%,and the non-aGVHD group after transplantation was(49.10+10.79)%,the aGVHD group after transplantation was(23.44±4.46)%.Compare the percentage of TSP-1 positive cells between pre transplant group(47.92±9.24)% and non-aGVHD group(49.10 + 10.79)% after transplantation,the difference was not statistically significant(P>0.05).But compare the percentage of TSP-1 positive cells between aGVHD group(23.44±4.46)% after transplantation and pre transplant group(47.92±9.24)%,The difference was statistically significant(P<0.05),compare aGVHD group(23.44±4.46)% after transplantation and non-aGVHD group(49.10 + 10.79)% after transplantation,The difference was statistically significant(P<0.05)too.the expression of TSP-1 in aGVHD group after transplantation was significantly lower than pro transplantation group and non-aGVHD group after transplantation.Conclusion:the expression of TSP-1 on the surface of dendritic cells in patients with acute aGVHD significantly lower than pro transplantation group and non-aGVHD group after transplantation,the expression of TSP-1 on dendritic cells was correlated with the occurrence of aGVHD.
