Objective: To explore the correlation between mast cell degranulation and myocardial cell injury by measuring the level of inflammatory mediator such as histamine,tryptase and complement in CE-induced allergy and further explore the possible mechanism.Methods: Balb/c mice were randomized into group A(healthy control),group B(E.g infection),and group C(DEX preconditioning).The mouse model of echinococcosis infection was established by microcapsule method in group B and group C.In the control group,0.9% normal saline was injected intraperitoneally,while in the other groups,peritoneal injection of hydatid cyst fluid was used to induce hypersensitivity to establish a mouse model of hydatid-induced allergic reaction.The severity of anaphylaxis in each group was compared using a comprehensive allergy assessment system.The levels of histamine and complement C3 a in serum were quantified by enzyme-linked immunosorbent assay,the levels of tryptase and cardiac troponin I in myocardial tissues were quantified by Western blot.Pathological sections of the apex of the heart were made and stained with HE,and the pathological changes were observed under the microscope.Results:(1)8 weeks later mice were dissected and found to be filled with cysts of different sizes.(2)Compared with group A,the levels of histamine and complement C3 a in group B and group C increased,and the levels of tryptase and cardiac troponin I increased P<0.05),but the above indicators in group C were lower than those in group B(P<0.05).(3)The scores of mice in group B and C were higher than those in group A,but the scores of mice in group B were higher than those in group C.(4)Compared with group A,the cardiomyocytes in group B and C showed different degrees of vacuolar denaturation cell infiltration and fibrous tissue proliferation,but the degree of damage in group C was less than that in group B.Conclusion:(1)In the allergic reaction caused by echinococcosis,mast cell degranulation releases a large amount of tryptase and HIS.These two substances can promote myocardial fiber rupture,myocardial fiber tissue proliferation,and intermuscular vasospasm and ischemia,thus leading to myocardial injury.(2)Dexamethasone can inhibit the release of histamine,trypsin and other inflammatory substances,thereby reducing the myocardial damage caused by allergic reactions to a certain extent.
