Objective : To study the effect of FBXW7 gene(also known as CDC4,FBW7 and h CDC4)on the biological behavior of human esophageal invasion and migration.Methods:Culture human esophageal squamous carcinoma cells Eca109 and EC9706,using two differently differentiated esophageal squamous carcinoma cell lines,SiRNA and full-length cDNA overexpression technology were used to knock down FBXW7,and FBXW7 was overexpressed.Transwell migration experiment was performed to understand the migration function of esophageal squamous cell carcinoma and Transwell invasion experiment was used to understand the invasion function of esophageal squamous cell carcinoma.To observe the changes of invasion and migration ability of ESCC,and compare the role of FBXW7 expressed at different levels in the invasion and migration of esophageal squamous cell carcinoma.Results:Cell scratch test results: The cell healing rate of Eca109 and EC970 cellin,the FBXW7 overexpression,was significantly higher.than the negative and blank control groups(P <0.05);Transwell migration: the number of cells permeating the membrane of FBXW7 overexpression group of ECa109 and EC9706 cells were(188.64(± 16.19)and(79.90 ± 6.24)were significantly increased compared with the corresponding control group(P <0.01);the number of cell membranes permeated by the siRNA FBXW7 group of ECa109 and EC9706 cells was(217.33 ± 45.99)and(232.42 ± 54.25)were correspondingly higher The control group decreased(P <0.01).Transwell invasion: ECa109,EC9706 cells FBXW7 overexpressing group cell transmembrane numbers were(162.36 ± 8.64),(72.50 ± 6.52)were significantly increased compared with the corresponding control group(P <0.01);ECa109,EC9706 cells siRNA FBXW7 group cells The number of perforated membranes were(57.85 ± 20.92)and(65.81 ± 18.12),which were significantly less than the corresponding control group of the number of perforating cells(P <0.01).Conclusion:When FBXW7 is overexpressed,it promotes the migration and invasion of ECa109 and EC9706 cells;FBXW7 siRNA inhibits the migration and invasion of ECa109,EC9706 cells.Different expression levels of FBXW7 have different ability to migrate and invade esophageal squamous cell carcinoma.FBXW7 facilitates the migration and intrusion of ESCC,It is suggested that FBXW7 is involved in the malignant progression of esophageal cancer.
