Abstract/Summary:
Objective: To study the effect of Xiaoyao Powder and simplified formula in reducing the liver toxicity of Tripterygium wilfordii,and further study the mechanism of reducing the liver toxicity of Tripterygium wilfordii.Methods: Methods: 1.Preparation of Tripterygium wilfordii model: 20 SD rats were divided into the blank group and the model group according to the method of random number table.The blank group was administrated with normal saline,and the model group was administrated with Tripterygium wilfordii Decoction of 2g/ml,lasting for 4 weeks.In the second and fourth weeks,blood was taken from orbit respectively,and the levels of ALT and AST were detected.The hepatotoxicity of Tripterygium Wilfordii in the first few weeks was studied.2.Comparison of Xiaoyao Powder and simplified formula in alleviating the liver toxicity of Tripterygium wilfordii: 60 SD rats were divided into blank group,model group,Xiaoyao Powder group,simplified formula group and Ganlixin group,12 rats in each group.Except blank group,the other groups were given Tripterygium wilfordii Decoction for 4 weeks.After that,the blank group and Tripterygium wilfordii group were infused with normal saline,the other groups were infused with corresponding treatment drugs.On the first day after the end of gavage,the experimental animals were taken,the serum of abdominal aorta was taken to measure the liver function indexes ALT and AST,the indexes related to oxidation GSH-Px,SOD and MDA,and the liver weight of rats was weighed to calculate the liver index.The same part of the liver was put into the fixed solution to make pathological sections,and the degree of pathological damage was calculated.3.The mechanism of Xiaoyao Powder to reduce the hepatotoxicity of Tripterygium wilfordii: the content of VEGF in serum was detected by ELISA,the right hypothalamus was put into the fixed solution to make pathological sections,and the protein expression levels of VEGF,BDNF and p-ERK2 in liver tissue were measured by Western blot.Results: 1.Compared with the blank group,the ALT and AST in the model group increased at the 2nd and 4th week,but the hepatotoxicity was more serious at the 4th week.2.Compared with the blank group,the ALT and AST levels in the Tripterygium wilfordii group increased significantly(P<0.01)Compared with Tripterygium wilfordii group,the ALT and AST levels of Xiaoyaosan group,simplified formula group and Ganlixin group were significantly lower(P<0.01)。 Compared with the blank group,the level of MDA in Tripterygium wilfordii group increased significantly((P<0.05)Compared with Tripterygium wilfordii group,the levels of Xiaoyaosan group,simplified formula group and Ganlixin group were significantly lower(P<0.05)。The content of SOD and GSH-Px in the model group was lower than that in the blank group(P<0.01)。Compared with Leigong group,Xiaoyaosan group increased significantly(P<0.01)。The liver index of Tripterygium wilfordii group was significantly higher than that of blank group(P<0.01)Compared with Lei Gong group,liver index of Xiaoyao Powder group,simplified formula group and Ganlixin group decreased significantly(P<0.01)。 Inflammatory cell infiltration appeared in the pathological section of Tripterygium wilfordii group,and the hepatocytes of Xiaoyao Powder group and Ganlixin group were almost normal.3.The VEGF level of the model group was higher than that of the blank group,while the VEGF level of the Xiaoyaosan group and Ganlixin group was significantly lower than that of the model group.Western blot results showed that the expression level of BDNF protein in the model group decreased,while the expression level of p-ERK2 and VEGF increased.Compared with the model group,the expression level of p-ERK2 and VEGF in the Xiaoyao Powder group,the simplified formula group and the Ganlixin group decreased.The pathological section of the hypothalamus showed that the proliferation of glial cells in the model group was active,the cells were slightly heteromorphic,and the hepatocytes in the simplified formula group and the Ganlixin group were generally normal.Conclusion: Tripterygium wilfordii can induce hepatotoxicity model when the dosage of Tripterygium wilfordii is 2g/ml.Xiaoyao Powder and simplified formula have liver protective effects,among which,Xiaoyao Powder has the best effect on reducing the liver toxicity of Tripterygium wilfordii.It is found that Xiaoyao Powder has the best effect,whether it is to reduce ALT and AST levels,or to improve the antioxidant level.The mechanism of Xiaoyao Powder to reduce the hepatotoxicity of Tripterygium wilfordii may be related to VEGF signaling pathway.
