Chronic kidney injury(CKI)is a relatively common refractory disease.As the incidence of chronic kidney disease has increased globally,it has become a global public health safety issue,which is currently not effective.How to delay the development of chronic kidney disease has become a research hotspot.With the increasing number of reports on the research and utilization of mushroom polysaccharides,the role of polysaccharides has been more and more recognized and paid attention to and because of its role in food therapy and health care,fungi have become increasingly important,showing a huge economy and social values.In this paper,enzymatic-hydrolysis Auricularia polytricha polysaccharide(EnAPS)was obtained from A.polytricha polysaccharide(APS),and its in vitro antioxidant capacity and monosaccharide composition were detected.The mice experiment results showed that EnAPS had the anti-oxidant,anti-inflammatory and anti-apoptotic effects on adenine-induced chronic kidney injury.The main results are as follows:(1)High performance liquid chromatography(HPLC)showed that EnAPS was mainly composed of mannose,ribose,glucuronic acid,glucose,galactose and rhamnose with mass fractions of 1.84%,0.27%,0.39%,24.7%,0.62% and1.90%,and the molar ratio was 10: 1.8: 1.7: 137: 3.4: 12.7.Fourier transform infrared spectroscopy(FT-IR)results indicated that EnAPS contained characteristic absorption peaks of polysaccharides,which were pyranose with ?-glycosidic bonds.(2)At 1000 mg/L,the scavenging effects of EnAPS on hydroxyl,superoxide anion and 1,1-diphenyl-2-trinitrophenylhydrazine(DPPH)radicals were 4.08 ± 0.15%,44.70 ± 1.69% and 112.00 ± 4.57%,respectively,which was 30.67%,8.84% and 11.31% higher than those of APS.The reducing power of EnAPS was 0.16 ± 0.01,which was 34.78% higher than that of APS.(3)Renal protective effects of EnAPS on adenine-induced chronic kidney injury of mice were investigated.EnAPS could reduce the content of creatinine(SCr),blood urea nitrogen(BUN)and increase the content of uric acid(UA)in model mice.It could increase the activity of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),catalase(CAT)in kidney tissue,and inhibit lipid peroxidation production of malondialdehyde(MDA).The levels of tumor necrosis factor-alpha(TNF-?),interleukin-6(IL-6)and phosphorylated nuclear factor kappa-B(p-NF-?B)were decreased.The qRT-PCR results showed that EnAPS could reduce the expression of apoptosis-promoting factor and Bax at the mRNA level,and increase the expression of apoptosis-inhibiting gene Bcl2,indicating that EnAPS might alleviate chronic kidney injury.The results of H&E sections,masson-stained sections and apoptotic sections all showed that,with the intervention of EnAPS,the inflammation and apoptosis of the kidney tissue were significantly repaired.The above results indicated that EnAPS could improve the anti-oxidative ability,reduce the inflammation,alleviate tissue fibrosis and apoptosis,and have a certain repair effect on adenine-induced chronic kidney injury.
