ObjectiveLaryngeal Squamous Cell Carcinoma(LSCC)is a common malignant tumor in the ENT department.In recent years,the incidence has been increasing.It is characterized by high malignancy,no obvious symptoms,early cervical lymph node metastasis,high recurrence rate,and severely affecting the quality of life of patients after surgery.It has been a challenge faced by clinicians.Finding potential diagnostic targets for laryngeal cancer in biomolecules has been a research hotspot in recent years.GRHL3,as a transcription factor,plays an important role in the development of a variety of cancers and normal human tissues,and there is not much research in the field of laryngeal cancer.Immunohistochemical staining was performed on postoperative laryngeal cancer tissue wax blocks,and human laryngeal cancer cell lines used RT-qPCR,western blot,cell transfection technology,and functional experiments to investigate the expression and biological behavior of GRHL3 in laryngeal cancer.It is hoped to provide certain ideas and foundation for the study of laryngeal cancer GRHL3.MethodsThrough routine pathological examination and immunohistochemical staining of postoperative pathological wax blocks and normal tissues adjacent to the cancer in patients with laryngeal cancer,human laryngeal cancer cell lines were analyzed by RT-qPCR and western blot to investigate the expression of GRHL3 in laryngeal cancer tissues and laryngeal cancer cells.Cell transfection technology,transwell experiment and CCK-8 experiment confirmed that silencing GRHL3 expression can reduce the migration,invasion and proliferation ability of laryngeal cancer cells,and can affect tumor cell formation epithelial-mesenchymal transition(EMT)The expression of the protein and whether it is related to the activation and regulation of the Wnt/?-catenin signaling pathway.These results suggest that GRHL3 may become a molecular target for research and diagnosis of laryngeal cancer in the future.Results1.40 cases of laryngeal squamous cell carcinoma after operation tumor tissue and paraffin normal tissue wax block immunohistochemistry showed:GRHL3 expression in laryngeal squamous cell carcinoma tissue was significantly higher than normal tissue,which was closely related to lymph node metastasis,clinical stage,and histological grade(p<0.05).2.Using transient cell transfection technology to silence the expression of GRHL3 can reduce the invasion,migration and proliferation capacity of human laryngeal cancer cells.3.Silencing GRHL3 expression can up-regulate E-cadherin,an important marker protein in EMT,and decrease Vimentin protein in tumor cells,and down-regulate?-catenin protein and cyclinD1 protein in the Wnt/?-catenin signaling pathway.Conclusions1.GRHL3 is closely related to the clinical and pathological parameters of patients with laryngeal squamous cell carcinoma.2.Silencing GRHL3 can reduce the invasion,migration and proliferation of human laryngeal cancer cells.3.The migration and invasion of laryngeal cancer ptomote EMT is closely related to GRHL3 and may be regulated by the Wnt/?-catenin signaling pathway.
