Objective: Initially explore the effect of dihydrotestosterone on hair and hair follicles in the healing process of burned mouse and select the appropriate concentration of dihydrotestosterone.The process of proliferation,differentiation and migration of hair follicle stem cells during the healing process of burns was detected by immunofluorescence labeling.Methods:In the body hair follicle culture,different concentrations of dihydrotestosterone were injected subcutaneously into the burned mouse,and the hair shafts were taken for scanning and observation.Using microsurgical dissection techniques,the wound skin was isolated,embedded in OCT,and HE stained to observe the number and morphology of hair follicles after injection of different concentrations of dihydrotestosterone.Using EdU and CD200 immunofluorescence staining to observe,the process of proliferation,differentiation and migration of hair follicle stem cells during the skin healing process of burned mouse after dihydrotestosterone injection.Results: Through experiments,it was found that different concentrations of androgen had no obvious effect on the hair shaft and hair skin during the healing process of burned mouse,but had an obvious effect on the hair follicles.Among the androgens in a certain concentration range,7.5mg/kg·d dihydrotestosterone was selected to inhibit the hair follicles in the skin healing process of burned mouse.EdU cell proliferation test found that 7.5 mg/kg·d dihydrotestosterone can inhibit the proliferation of cells and make the hair follicles shrink.Stem cell marker CD200 immunofluorescence showed that CD200-positive hair follicle stem cells were mainly concentrated in the outer root sheath bulge of hair follicle;At the same time,7.5 mg/kg · d dihydrotestosterone can inhibit the expression of CD200 positive hair follicle stem cells.Conclusion: A certain concentration of dihydrotestosterone can inhibit the hair follicles in the healing process of burned mouse,so that the hair follicles during the healing period are reduced,and the hair follicles become thin and atrophy.At the same time,a certain concentration of dihydrotestosterone can inhibit the expression of CD200 positive hair follicle stem cells in the outer root sheath bulge of the hair follicle.
