Objective:To investigate the effect of the molecular targeted drug imatinib mesylate on proliferation,apoptosis and migration in human colorectal cancer cell line SW-837.Methods:(1)Cultured human colorectal cancer cell line SW-837 in vitro.CCK-8 assay was applied to detect the impact on proliferation of SW-837 cells treated with different concentrations of imatinib mesylate for 24 hours.(2)Annexin V-EGFP,PI double staining and flow cytometry were used to detect the apoptosis of SW-837 cells treated with different concentrations of imatinib mesylate for 24 hours.(3)Cell scratch experiment was used to observe the migration of SW-837 cells treated with different concentrations of imatinib mesylate for 48 hours.Results:(1)Using an inverted microscope to observe SW-837 cells,the cells of control group grew well.However,the cell proliferation speed of the groups which were treated with different concentrations of imatinib mesylate,was apparently slowed and the gap between cells became lager.Meanwhile cells detached from the wall and debris was visible.With the increase of drug concentration,this effect was more obvious.(2)CCK-8results showed that compared with the control group,when the imatinib mesylate concentration in the experimental group was 5-80?mol/L,it had an inhibitory effect on the proliferation of SW-837 cells and theinhibition rate was 27.2%-61.3%.With the increase of drug concentration,the inhibition rate increased gradually(P<0.01).(3)The results of flow cytometry showed that the apoptosis rate of the groups which treated with different concentrations imatinib mesylate was significantly higher than the blank control group(P<0.01).With the increase of drug concentration,the apoptosis rate increased gradually(P<0.01).(4)Cell scratch test showed that the groups which treated with imatinib mesylate and with the concentration over 20?mol/L,had lower healing rate than the blank control group(P<0.01).It was suggested that imatinib mesylate could inhibit the migration of SW-837 cells.But with the increase of drug concentration,the healing rate decreased gradually(P<0.01).Conclusions:(1)Imatinib mesylate inhibited the proliferation of SW-837 cells in a dose-dependent manner.(2)Imatinib mesylate can effectively induce the apoptosis of SW-837 cells in a dose-dependent manner.(3)Imatinib mesylate inhibited the migration of SW-837 cells in a dose-dependent manner.
