Objective:To evaluate the therapeutic effect of tacrolimus by applying donor derived plasma cell-free DNA in kidney allograft recipients.And to reexamine minimally effective trough tacrolimus(Tacro)concentrations in renal transplantation patients.Materials and Methods: Retrospectively collected 14 patients with stable allograft who were 15 days after renal transplantation and were regularly access to the immunosuppressive therapy of tacrolimus including 4 females(age=34.00±7.94)and 10 males(age=35.67±8.32).Both plasma ddcf DNA level and Tacro concentrations measured.Samples were analyzed using droplet digital polymerase chain reaction for ddcf DNA and liquid chromatography tandem mass spectrometry for Tacro.ddcf DNA and trough Tacro concentrations were then compared to identify Tacro concentrations associated with intact graft integrity.Results: the tacrolimus therapy applied in our center is sufficient and effective.But cannot find out the minimum concentration of tacrolimus after renal transplantation.
