Objective Pressure ulcers(PU)is one of the most common complications in ICU and long-term bedridden patients.This project intends to use rat pressure ulcer model to prepare Autologous Platelet Rich Fibrin gel and observe its therapeutic effect on rat pressure ulcer healing.At the same time in vitro cell and tissue immunohistochemistry experiments were used to explore its possible in vivo and in vitro mechanisms which laid a theoretical and experimental foundation for the clinical application of this technique in the treatmnet fo pressure ulcers.Methods(1)rat pressure ulcer model was prepared by using the principle of ischemia-reperfusion injury.(2)venous blood of pressure ulcer model rats was collected to prepare Autologous Platelet Rich Fibrin gel and its therapeutic effect on pressure ulcer healing in rats was observed.(3)preparation of 1640 medium containing Autologous Platelet Rich Fibrin leach solution.(4)in vitro cell proliferation Transwell and scratch healing tests were used to analyze the effects of Autologous Platelet Rich Fibrin gel on the proliferation and migration of vascular endothelial cells.The expression of VEGF in vascular endothelial cells and pressure ulcer tissues was analyzed by WB and IHC.Results(1)Using the principle of "ischemia-reperfusion injury" the rat pressure ulcer model was successfully prepared to simulate the clinical pressure ulcer.Subsequently Autologous Platelet Rich Fibrin gel film was used to prepare rat pressure ulcer model.The results showed that Autologous Platelet Rich Fibrin gel could significantly promote the wound healing of rat pressure ulcer.(2)The in vitro cell experiment results showed that the medium containing leached solution of Autologous Platelet Rich Fibrin gel could significantly promote the proliferation and migration of vascular endothelial cells.(3)HE staining results of pressure ulcer tissues showed that compared with the control group the Autologous Platelet Rich Fibrin gel treated pressure ulcer tissues were basically intact after the seventh day.WB and IHC results showed that compared with the control group the expression of VEGF in the pressure ulcer tissues and vascular endothelial cells significantly increased after Autologous Platelet Rich Fibrin gel treatment.Conclusion Autologous Platelet Rich Fibrin gel can promote the healing of pressure ulcer and its possible mechanism is that it can promote the proliferation and migration of vascular endothelial cells and promote the expression of VEGF in vascular endothelial cells and pressure ulcer tissues.
