Malignant tumor is a serious threat to human health.The research on the occurrence and prevention of tumor is still the focus of life science.Uveal Melanoma(UVM)is one of the most common intraocular malignancies in adults.The incidence of this disease is the first and the second most common intraocular malignancy in foreign and domestic patients.It is very prone to blood flow metastasis and has a high fatality rate.As for the treatment of UVM,at present,the main means of eyeball resection is eye resection.In order to preserve the eyeball and partial vision of the patients,various conservative treatment methods,such as phototherapy,radiation therapy,proximity radiotherapy and immunotherapy,have been developed,but the effect is not ideal and further improvement is needed.Therefore,it is of great significance to explore new techniques and methods of UVM gene therapy.Objective:To study the expression of Calcium and integrin binding protein-1(CIB1)in UVM disease and its significance,provide a new method for guiding the treatment of UVM melanoma,evaluating the prognosis of UVM patients and preventing and treating UVM.Method:Using Gene Expression data interaction Analysis GEPIA(Gene Expression Profiling Interactive Analysis)database,cancer chip database(UALCAN)these online database Analysis CIB1 differentially expressed in uveal melanoma organizations,CIB1 in uveal melanoma tissues Expression level corresponding relationship with the prognosis and the correlation with CIB1 factors associated factors.CBioPortal was used to analyze the mutation of CIB1 in uveal melanoma and other tissues.The protein-protein interaction network(PPI)was further constructed by STRING database.In addition,we also used the comprehensive analysis of STRING and GeneMANIA databases to predict cibl-related genes in UVM,and verified the similarity between gene expression and survival heat map in UVM.Finally,we collected the uveal tissues and uveal melanoma tissues of the normal eye,respectively,and performed CIB1 factor immunohistochemical staining and pathological examination of the two groups.Results:Through immunohistochemical detection,we found that compared with normal uveal tissue samples,the expression of CIB1 in tumor tissues of evanescent melanoma was significantly enhanced(P<0.05).The expression of CIB1 in uveal melanoma was independent of weight and gender.However,the expression of CIB1 was related to the stage of uveal melanoma,the age of uveal melanoma patients and the pathological grade of uveal melanoma patients(P<0.05).Moreover,the high expression of CIB1 significantly reduced the survival rate of patients with evanescent melanoma(P<0.05).At the same time,the study data showed that the expression of CIB1 in grape melanoma tissues was related to its grade,stage,race and gender,especially the expression level of CIB1 was significantly higher in the fourth stage of tumor classification than in the third stage.The results suggested that CIB1 and the genes it interacts with are significantly enriched in ITGA2B-ITGB3-CIB1 complex,regulation of intracellular protein transport,regulation of ion transport,cellular response to tumor necrosis factor and positive regulation of apoptotic process.cBioPortal for Cancer Genomic analysis demonstrated that there are no mutations have been detected in the current TCGA-UVM samples,but several gain,dioploid and shallow deletion were seen in patients with UVM.Methylation analysis revealed that stage,age and gender of uveal melanoma patients may have little effect on CIB1 promoter methylation in UVM patients.However,body weight may alter promoter methylation levels in UVM patients.PPI analysis suggested that CIB1 has a greater interaction with 11 factors such as GUF1,CRY2,PLK3,ACTN4,PSEN2,ITGA2B,ITGB3,MAP3K5,CABP1,SPICE1,and ARHGEF1.Conclusion:To sum up,our new findings suggest that CIB1 may be a potential diagnostic predictor of UVM and a potential target for the genetic diagnosis,treatment and prognosis of melanoma of the grape.
