| Objective:Gastric cancer is a common cancer with high mortality and low survival rate,and bioinformatics methods to dig out its susceptible genes are currently more commonly used analysis tools.In addition,immunotherapy is a new and effective cancer treatment method to analyze its immune cells.Invasion can help clinical implementation of gastric cancer immunotherapy.This study screened genes susceptible to gastric cancer in the population in the Middle East and Asia,and aimed to understand the pathogenesis of gastric cancer from a genetic perspective,so as to help the diagnosis and treatment of gastric cancer.In addition,this study also analyzes the immune infiltrating cells of gastric adenocarcinoma,in order to find the immunotherapy target of gastric adenocarcinoma from the cytology point of view,and provide a more reliable basis for choosing a more appropriate treatment.Methods :(1)Search in the public database GEO,use the GEO2 R platform to screen for differential genes in gastric cancer,perform GO enrichment analysis and KEGG signal transduction pathway analysis,and use STRING to construct the association of gene-related protein interactions.(2)Search the gastric adenocarcinoma cases in the public database TCGA,and use Perl script to organize the data.The study of gastric adenocarcinoma immune invasion included a total of 348 patients,including 343 cancer samples and 30 normal samples.The CIBERSORT deconvolution algorithm was used to evaluate the proportion of different immune cells in tumor tissues.Finally,R software was used for differential immune cell analysis,clinical Correlation analysis and survival analysis.Results:(1)Two data sets were included in the study of differential genes.Through screening,40 differential genes were obtained,of which 34 were up-regulated genes and 6 were down-regulated genes.ARHGAP11 A gene was found to be associated with gastric cancer for the first time.Analysis of the differential genes revealed that the signaling pathways involved mainly included leukocyte transporter migration,p53 signaling pathway,and cell cycle.(2)The results of immune invasion of gastric adenocarcinoma showed that the inactivated CD4 memory T cells,CD8 T cells,M2 macrophages and M0 macrophages in tumor samples had the most obvious immune infiltration,accounting for 15.90% and 12.67%,respectively,11.81%,and 12.05%.(3)Differential analysis of immune invasion in gastric adenocarcinoma showed that plasma cells,monocytes,activated CD4 memory T cells,M0 macrophages,M1 macrophages and M2 macrophages were different in normal samples and tumor samples(P < 0.05).(4)The clinical correlation of immune infiltration of gastric adenocarcinoma showed that the inactivation of unactivated dendritic cells and CD8 T cells increased with the increase of gastric adenocarcinoma classification,showing a positive correlation(P <0.05).T cells,M0 macrophages,active mast cells,and neutrophils showed a negative correlation with grading(P <0.05).(5)Gastric adenocarcinoma immune infiltration survival analysis found that unactivated CD4 T cells had a negative correlation with five-year survival rate(P = 0.029).CD8 T cells were positively correlated with the five-year survival rate(P = 0.002).Conclusions:(1)The bioinformatics method can effectively screen out gastric cancer susceptibility genes in the population in the Middle East,so as to provide a basis for the study of the pathogenesis of gastric cancer.(2)The CIBERSORT method can effectively screen the proportion of gastric adenocarcinoma immune cells.Unactivated CD4 memory T cells,CD8 T cells,M2 macrophages and M0 macrophages play an important role in gastric invasion and prognosis.The immune cells with more immune infiltration can be used as drug targets to improve the survival rate of patients. |
