| Objective: To analyze whether HOTAIR and hsa-miR-148b-3p are bound by bioinformatics,and then verify the interaction between the two in breast cancer cell lines through cytological experiments,and provide directions for the next step to explore the regulatory mechanism of the two.Methods: In this study,high-throughput miRNA data was downloaded from the TCGA database.According to cut-off criteria(P <0.05,log2 FC> 2.0),a total of 112 differentially expressed miRNAs were detected in breast tumors and normal specimens.HOTAIR target miRNAs were screened from the miRcode database,including 55 miRNAs including miRNA-148b-3p.Based on the Veen diagram,the two groups of data were identified as overlapping genes,namely miR-148b-3p,miR-204-5p,and miR-375.We used Kaplan-Meier curve and log rank sum test to analyze the survival of the above three miRNAs.The results showed that miR-148b-3p and miR-204-5p had significant effects on the survival of breast cancer patients,while miR-375 had no significant difference.Furthermore,in the starBase database we noticed that only miRNA-148b-3p and the hyperconserved region of HOTAIR showed a complementarity of> 10 bases.Double luciferase experiments were used to verify the binding of HOTAIR-miRNA-148b-3p and miRNA-148b-3p-ESR1 in vitro.Another cytological experiment further verified that the three candidates had 15 breast cancer samples and breast cancer cell line MCF-7.Analysis of expression,and analysis of expression correlation.Results: Veen diagram was used to screen the differentially expressed miRNAs and HOTAIR target miRNAs for overlapping data screening,and then survival analysis was performed.Finally,miR-148b-3p and miR-204-5p were found to have significant expression differencesin breast cancer specimens and to breast The survival of cancer patients has a significant impact,however in the starBase database we found that only miRNA-148b-3p binds to HOTAIR.The double luciferase assay HOTAIR-miRNA-148b-3p can all bind in vitro.Cytological experiments have confirmed that high expression of HOTAIR can inhibit miRNA-148b-3p expression in breast cancer.Conclusion: HOTAIR may interfere with the expression of miRNA-148b-3p as a competitive endogenous RNA(ceRNA),and then regulate downstream genes,thereby affecting the occurrence and development of breast cancer. |
