Objective:To investigate the correlation between cervical indoleamine 2,3-dioxygenase and HPV E6/E7 protein and cervical cancer.Methods:From September 2016 to December 2017,patients with cervical lesions were selected for the first time in gynecology department of the Third Affiliated Hospital of Kunming Medical University,by means of HC-2 to exame High risk HPV 30 cases of patients with pathological diagnosis of cervical cancer as the experimental group,15 cases of no cervical lesions of complete hysterectomy women as the control group Collected in the experimental group of patients with tumor tissue and control group of cervical tissue,the collected specimens were preserved by polyformaldehyde.Adopt immunohistochemical staining to exame indoleamine 2,3-dioxygenase and HPV E6/E7 protein.To explore the possible interaction between IDO and HPV E6/E7 protein in the progression of cervical cancer.Results:1.Bar graph show that IDO expression and HPVE6/E7 protein in cervical cancer and normal cervical tissue samples by IHC.The rank sum test of two independent samples was used.The IDO,E6 and E7 values of Mann-Whitney U are 45.00,60.00 and 7.50,respectively,the expression of IDO,E6 and E7 in cervical cancer group was higher than that in normal group,P value was<0.001(Table 3.Fig.2).The percentage of positivity increased in relation to the increment of disease stage:15%,30%for stage?,?,respectively in IDO experssion,the percentage of IDO in stage ? was 10.00%,and that in Phase ? was 5.00%because of the fewer cases in stage ? and ?(Table 4,Fig.3).15%,35%for stage ?,?,respectively in HPV E6,the percentage of HPV E6 in stage ? was 10.00%,and that in Phase ? was 5.00%because of the fewer cases in stage ? and ?(Table 5,Fig.4).20%,55%for stage ?,?,respectively in HPV E7,the percentage of HPV E7 in stage ? was 15.00%,and that in Phase ? was 5.00%because of the fewer cases in stage ? and ?(Table 6,Fig.5).2.According to the receiver operating characteristic curves(ROC),diagnostic ROC Curve of Myometrial Infiltration>1/2,when the sensitivity of HPV E6 was 0.667,the specificity was the highest:0.333.The area under the curve of E6 is 0.648,the area under the curve of E7 is 0.444,and the area under the curve of IDO is 0.593.The area under the curve of E6 is the largest and the diagnostic efficiency is the highest.ROC curve for diagnosis of lymph node metastasis,when the sensitivity of IDO experssion was 0.800,the specificity was the highest:0.714.The area under the curve of IDO is 0.629,the area under the curve of E7 is 0.400,and the area under the curve of IDO is 0.557.The area under the curve of IDO is the largest and the diagnostic efficiency is the highest.3.The results of box plot that the 95%confidence interval overlaps,P>0.05,and there is no significant difference among the IDO expression and HPVE6/E7 protein in cervical cancer and normal cervical tissue samples by IHC.4.The results of rank sum test showed that according to the results of immunohistochemical staining,HPV E6 protein,HPV E7 protein and IDO were used to analyze cervical squamous cell carcinoma:?2(H)=3.808,v=2,P=0.149,and there was no significant difference among the three methods for cervical adenocarcinoma:x2(H)=0.876.v=2,P=0.645.with no significant difference.5.Chi-square test analysis:In different differentiated cervical cancer tissues,HPV E6/E7 protein and IDO were detected by immunohistochemical staining,?2=4.659,P=0.995.with no significant difference.The results showed that the positive rates of HPV E6/E7 protein and IDO were basically the same.HPV E6/E7 protein.IDO,?2=3.183,P=0.672 were detected by immunohistochemical staining in cervical cancer tissues infiltrated by different muscle layers,with no significant difference;the results showed that the positive rates of HPV E6/E7 protein and IDO were basically the same;the lymph node metastasis in cervical cancer tissue was detected by immunohistochemical staining for HPV E6/E7 protein,IDO,?2=1.810,P=0.875,with no significant difference.The positive rates of HPV E6/E7 protein and IDO in detecting lymph node metastasis were basically the same.Conclusions:1.The ROC curve of diagnosis with or without lymph node metastasis shows that the area under the curve of IDO is the largest,the diagnostic efficiency is the highest,the expression of IDO is high,and the possibility of lymph node metastasis is high;the ROC curve of muscle invasion>1/2 or not shows that the area under the curve of HPV E6 is the largest,the diagnostic efficiency is the highest,HPV E6 is high,and the possibility of muscle invasion is high.2.The 95% confidence intervals of IDO expression,HPV E6 and HPV E7 were coincident,P>0.05.There was no significant difference among them.The expression of IDO,HPV E6 and HPV E7 in cervical cancer tissues was basically the same.The combination of them could be used to monitor the occurrence,development and prognosis of cervical cancer.3.The positive rates of IDO and HPV E6/E7 protein expression in different cervical histopathology,differentiation,stage,muscle invasion and lymph node metastasis of cervical cancer were basically the same.
