Objectives:This study was designed to research the influence of diatom contamination in the results of diatom detection,and the significance of conventional cleaning in diatom detection,to explore the applicability of diatom detection in the case of experimental materials contamination.Methods:1.The content of diatom in lungs,livers,kidneys of drowning cases were tested by the method of the combination of microwave digestion,vacuum filtration and scanning electron microscope,finding the ratio of the content of diatom in lungs and water samples.2.Water samples collected from the drowning site and tap water were used as pollution sources,finding the ratio of the content of diatom in lungs and water samples.2.1 The experimental group of drowning was divided into contaminated group and non-contaminated group:1)Take three portions(about 3g each)of liver tissue and kidney tissue respectively and one portion(about 1g)of lung tissue in the non-contaminated group.2)The contaminated group was divided into water sample contaminated group,water sample contamination cleaning group,tap water contaminated group and tap water contamination cleaning group.Take ten portions of lung tissue,liver tissue and kidney tissue respectively from each group,totally 120 portions.Each portion of liver tissue and kidney tissue was about 2g.Each portion of lung tissue was about 1g.2.2 The experimental group of non-drowning was divided into contaminated group and non-contaminated group:1)Take three portions(about 3g each)of liver tissue and kidney tissue respectively and one portion(about 1g)of lung tissue in the non-contaminated group.2)The contaminated group was divided into water sample contaminated group,water sample contamination cleaning group,tap water contaminated group and tap water contamination cleaning group.Take ten portions of lung tissue,liver tissue and kidney tissue respectively from each group,totally 120 portions.Each portion of liver tissue and kidney tissue was about 2g.Each portion of lung tissue was about 1g.2.3 To detect the content of diatom in each group of taken lung tissue,liver tissue and kidney tissue.Comparing the content of diatom in the same contaminated tissue between different pollution sources.Comparing the content of diatom between different contaminated tissue through the same pollution source.Comparing the content of diatom and the ratio between contamination and cleaning in each group.All the statistics were calculated by spss23.0.Results:1.The content of diatom in lung tissue of drowning cases was41989.06 ±25768.86/10g.The content of diatom in liver tissue was 10.25±6.78/10g.The content of diatom in kidney tissue was 18.44±14.76/10g.The ratio of the content of diatom in lungs and water samples was 25.15±21.07.2.The content of diatom in water samples is 25636/10mL.The content of diatom in tap water is 101/10mL.In experimental drowning group,the content of diatom in lung tissue of non-contaminated group was 65,952/1 Og.The content of diatom in liver tissue was 2/10 g.The content of diatom in kidney tissue was 1/10 g.The ratio of the content of diatom in lungs and water samples was 2.57.3.Drowning group:The average value of the content of diatom in lung tissue contaminated by water samples was 71079.94±26868.21/10g,and the average value of the content of diatom in liver tissue was 1403.04±1051.46/10g,and the average value of the content of diatom kidney tissue was 1642.68±1382.40/10g.The average value of the content of diatom in lung tissue contaminated by tap water was 69149.10±33565.10/10g,and the average value of the content of diatom in liver tissue was 857.89±724.67/10g,and the average value of the content of diatom kidney tissue was 329.90±278.33/10g.4.Non-drowning group:The average value of the content of diatom in lung tissue contaminated by water samples was 6885.22±4588.09/10g,the average value of the content of diatom in liver tissue was 827.67±458.60/10g,the average value of the content of diatom kidney tissue was 267.60±155.10/10g.The content of diatom in lung tissue contaminated by tap water was 190.18±174.05/10g,the average value of the content of diatom in liver tissue was 290.07±107.27/10g,the average value of the content of diatom kidney tissue was 412.15±191.55/10g.5.Drowning group after cleaning:The content of diatom in lung tissue contaminated by water samples was 67346.43 ± 15003.15/10g,and the content of diatom in liver tissue was 114.70 ± 106.82/10g,and the content of diatom in kidney tissue was 47.57±41.39/10g.The content of diatom in lung tissue contaminated by tap water was 66086.36 ± 30230.58/10g,and the content of diatom in liver tissue was 7.97 ± 6.62/10g,and the content of diatom in kidney tissue was 60.69 ± 54.33/10g.6.Non-drowning group after cleaning:The content of diatom in lung tissue contaminated by water samples was 55.71±50.35/10g,and the content of diatom in liver tissue was 11.30±8.70/10g,and the content of diatom in kidney tissue was 68.75 ± 56.83/10g.The content of diatom in lung tissue contaminated by tap water was 19.17±17.10/10g,and the content of diatom in liver tissue was 0/10g,and the content of diatom in kidney tissue was 33.86 ± 29.98/10g.7.The ratio of the content of diatom in lungs and water samples of four non-drowning contaminated groups all lower than 0.01 except the water sample contaminated group.Conclusions:1.Based on the method of filter enrichment in testing diatom,the detection rate of diatom in lung and kidney and liver tissue was 100%.2.The content of diatom in liver and kidney tissue was similar in drowning cases,and much lower than that in lung tissue.3.The contamination has little effect on the content of diatom in lung tissue of drowning cases.4.Cleaning is effective for diatom contamination.It should be cleaned by conventional ultrapure water in actual inspection.5.In the case of contamination,the ratio of the content of diatom in lungs and water samples was much lower than 2 in non-drowning cases.When the ratio was over 2,it was a valid evidence to support the diagnosis of drowning.
