Objective:To investigate the role of LNK-mediated JAK-STAT signaling pathway in hypertension vascular remodeling.Method:Rat thoracic aorta smooth muscle cells were selected to construct a hypertensive cell model.The experiment was divided into 4 groups:control group,HP group,no-load group,LNK group.Flow cytometry was used to detect the levels of VSMC cycle,apoptosis and proliferation;real-time quantitative PCR was used to detect the mRNA expression levels of LNK and JAK-STAT signal pathways in rat smooth muscle cells;Western blot was used to detect LNK,STAT3,p-STAT3,JAK2,p-JAK2 protein expression levels.Results:(1)The levels of vascular smooth muscle cell cycle,apoptosis and proliferation in each group of rats: Compared with the control group,VSMCs in the HP group and the no-load group decreased in the G0/G1 phase,increased in the S phase,and increased in the G2/M phase.There was statistical significance(P<0.05);compared with the no-load group,VSMC in LNK group increased in G0/G1 phase,decreased in S phase,and decreased in G2/M phase,the difference was statistically significant(P<0.05);VSMC in LNK group The apoptotic rate was higher than that of HP group and no-load group(P<0.05);the proliferation level of VSMC cells in HP and LNK groups was higher than that in control group(P<0.05);(2)mRNA expressions of LNK and JAK-STAT signal pathways on vascular smooth muscle cells of rats in each group: The expression levels of STAT3 and JAK2 mRNA in the HP group were significantly higher than those in the control group(both P<0.05);STAT3 and JAK2 in the LNK group The expression level of mRNA was significantly lower than that in no-load group and HP group(both P<0.05).(3)Expressions of LNK and JAK-STAT signaling pathway proteins on vascular smooth muscle cells of rats in each group: LNK protein expression levels in the HP group and the no-load group were significantly lower than those in the control group(P<0.05);the HP and no-load groups The protein expression levels of p-STAT3 andp-JAK2 were significantly higher in the control group(P<0.05);the protein expression levels of p-STAT3 and p-JAK2 in the LNK group were significantly lower than those in the no-load group of the HP group(P<0.05).Conclusion : LNcK may participate in vascular remodeling of hypertension by negatively regulating JAK-STAT signaling pathway.
