| Objective:to investigate the distribution of gene copy number variation(Copynumbervariations,CNV)in breast cancer under different states of Her-2,and to screen the key genes affecting Her-2 positive breast cancer,so as to lay a scientific basis for the treatment of Her-2 positive breast cancer.Methods:30 samples of breast cancer and paracancerous tissues from January 1,2018 to December 31,2018 were selected.The CNV of DNA was detected by whole exon high throughput sequencing,and the CNV of breast cancer in different states of Her-2 was analyzed.GO database,KE GGP AT H WAY database and protein-protein interaction(PPI)model were used to screen the core genes affecting Her-2 positive breast cancer.Results:in 30 cases of breast cancer and paracancerous tissues,there were 5 96 differences in the number of CNV between cancer and paracancerous tissues in CNV,Her-2 positive group and in Her-2 negative group.There was significant difference in CNV,between the two groups(χ2=21.10,P<0.01).In Her-2 positive group,there were frequent CNV amplification on chromosomes 1 and 8,the highest deletion region on chromosome 11 and a high heterozygous region on chromosome 17.In Her-2 negative group,chromosome 1 had a higher amplification region and chromosome 11 had a higher heterozygous region.Through comparative enrichment analysis,it was found that significant changes occurred in biological functions such as cell keratinization,oxygen transport capacity and cytochemical stress response,among which AKT1,VEGFA,ACTR1A,CAPN6 and EFTUD2 may be the key genes affecting Her-2 positive breast cancer.Conclusion:Her-2 positive CNV is more frequent than Her-2 negative CNV,and there are the most CNV on chromosomes 1,8,11 and 17.AKT1,VEGFA,ACTR1A,CAPN6 and EFTUD2 may play an important role in Her-2 positive breast cancer. |
