Prostatitis is a common disease that affects men's health,especially its recurring episodes that are difficult to heal are a major problem in treatment.The blood-prostate barrier makes it difficult for the drug to reach an effective concentration at the site of prostate disease,which is one of the main reasons that affect its efficacy.Researchers have always paid close attention to this.This subject takes the prostate as the research object,the drug concentration of the prostate tissue as the inspection target,and bibliometric analysis to understand the research progress in this field,and then combines the microdialysis technology and LC-MS detection method to carry out animal experimental research to discuss the blood-prostate barrier The mechanism and law provide a research basis for effective medical treatment of clinical prostate diseases.OBJECTIVE:To carry out literature quantitative analysis of the research on the topic of antibacterial drugs for prostatitis,and then to understand its research status,development trend and research difficulties;the microdialysis technique was used to determine the in vitro recovery rate and influencing factors of the antibacterial drug cefradine in order to optimize the method conditions for in vivo pharmacokinetic studies;the application of optimized sampling methods and measurement conditions to study the drugs in the blood and prostate tissues of Beagle dogs Kinetic changes,which laid a preliminary foundation for the study of the blood-prostate barrier.METHODS:1.Bibliometric analysis uses the CNKI database as the search target.It searches the literature related to the research on antibacterial drugs for prostatitis published in the database for a total of 27 years from 1993 to 2019 for visual analysis,and forms a related visual scientific network map.2.In vitro,use microdialysis concentration difference method(decrement method,increment method)and LC-MS to determine the in vitro recovery rate of cefradine,and investigate the influence of flow rate and concentration on the recovery rate to determine the use of microdialysis technology for cefradine In vivo recovery rate and conditions for pharmacokinetic studies.3.The recovery rate of the animal is determined with the determined microdialysis flow rate and standard concentration in the animal to determine the recovery rate in the body;then the animal pharmacokinetic experiment is carried out,the research is performed with the determined microdialysis flow rate and the drug concentration,the dialysate is collected and measured The drug concentration is finally converted to the actual concentration using the in vivo recovery rate.RESULTS:1.Bibliometric analysis found that the research on antibacterial drugs for prostatitis is still hot,mainly focusing on the prevention and treatment of prostatitis infection,selection of antibacterial drugs,clinical efficacy evaluation,etc.The use of antibacterial drugs has become an indispensable method for the treatment of prostate infections.Common antibacterial drugs include quinolones,cephalosporins and penicillins.2.The established LC-MS analysis method has good linearity within the required range,and the method is sensitive and reliable.There was no significant difference in the recovery rate measured by the increase-decrease method.Under the same conditions,the recovery rate of the probe in vitro decreases with the increase of the flow rate,and is not affected by the drug concentration around the probe.3.The pharmacokinetic results after intravenous administration show that the blood concentration of cefradine in normal beagle dogs is higher than that of the prostate,the peak concentration is equivalent to 38% of the peak blood concentration,its absorption and distribution is faster,and the half-life short.CONCLUSION:The treatment of prostatitis is still a hot and difficult issue,and it is inseparable from the use of antibacterial drugs;however,there are certain difficulties in the selection and dosage of antibacterial drugs.The reduction method can be used to determine the recovery rate of cefradine microdialysis in vivo,and the microdialysis technique can be well used to study the pharmacokinetics of the antibacterial drug cefradine in blood and prostate.
