OBJECTIVE:To verify the efficacy of Jia-Wei-Qing-Luo-Yin?JWQLY?on collagen-induced arthritis in rats and explore related mechanism of JWQLY,the optimized prescription of Qing-Luo-Yin?QLY?.METHODS:Thirty male SD rats were randomly selected,and divided into five groups on average,with one group as the control group and the other four groups as the treatment groups.The experimental group was modeled with bovine type?collagen.After modeling,3 groups were treated with QLY,JWQLY and methotrexate?MTX?,and the remaining group was retained as the model control group.The CIA rats were comprehensively evaluated and analyzed for a number of related indicators,including immunohistochemical indicators,general physiological indicators,histopathological indicators,morphological indicators,serum biomarker levels,etc.RESULTS:?1?Of general physiological indicators,the physiological status of rats in the treatment group was better than that of rats in the CIA model group;?2?of morphological indicators,the inflammatory index and swelling degree of paw joints in the treatment group were lower than those of rats in the CIA model group;?3?of histopathological indicators,QLY and JWQLY could reduce synovial inflammatory infiltration,improve joint space narrowing,improve joint bone destruction,inhibit synovial tissue proliferation;also JWQLY3 were more advantageous than MTX in terms of joint protection;?4?of histopathological indicators,QLY,JWQLY and MTX could effectively inhibit the expression of P-P65 protein in synovial tissue;?5?of serum biomarkers,QLY,JWQLY and MTX could effectively reduce the levels of inflammatory markers TNF-?,inhibit the expression of angiogenic factors VEGF,COX-2,inhibit PI3K/AKT and SphK/S1P signaling pathway,and for immune regulation,QLY and MTX could effectively inhibit IL-17A expression,and JWQLY could concurrently upregulate IL-35 expression and inhibit IL-17A expression.CONCLUSION:JWQLY embraces a good efficacy on arthritis in CIA model rats and can effectively improve the pathological changes of joints in CIA rats,which is helpful for the treatment of RA;the mechanism of JWQLY may be reducing the level of TNF-?in vivo,regulating the balance of Th17/Treg,reducing the level of angiogenic factors VEGF and COX-2 in vivo,inhibiting the multi-target effect of PI3K/AKT,NF-?B,SphK/S1P signaling pathway in vivo,relieving joint inflammation of CIA rats,and protecting the joints of CIA rats;JWQLY is with more advantages in immune regulation and angiogenesis compared with QLY;the formula idea of JWQLY provides a new idea for the optimization of clinical traditional Chinese medicine.
