| Objective:To study the anti-acute liver injury effect of persimmon parasitism,and screen the main active fractions,and preliminary study the anti-inflammatory mechanism on CC14 and APAP model.Methods:1)PP was prepared for concentrates by adding 80%alcohol and refluxing and extracting,extracting the concentrates used petroleum ether,ethyl-acetate,n-butyl alcohol.And get petrol ether extract,ethyl-acetate extract,n-butyl alcohol extract of PP.Mice was intragastrically treated with active part of PP for 8 days before CCl4 oil solution injection except for normal group.Serum levels of alanine aminotransferase(ALT),aspartate aminotransferase(AST)and the liver of biochemical indicators and hepatic histopathology was detected routinely.2)In brief,a total of 60 Kunming mice were divided into the control group,model group,bidentate positive group,and low,middle and high dose groups,Mice were intragastrically treated with active part of PP for 8 days before CCl4 oil solution injection except for normal group.Serum levels of alanine ALT,AST and the liver of biochemical indicators and hepatic histopathology was detected routinely.Using immunohistochemical technique and Enzyme-linked immunosorbent assay(ELISA)in hepatic tissue and serum of TGF-β1(transforming growth factor β1),TNF-α(tumor necrosis factor a),IL-6,IL-1β in the quantitative analysis.The IL-1β,JNK-MAPK,P38-MAPK,TNF-α gene and protein expression of hepatic tissue by Quantitative Real-time PCR and Western Blot the method to carry on the quantitative analysis.3)60 mice were divided into 6 groups randomly:normal control,model group,bidentate positive group,high-dose group,middle-dose group and low-dose groups.Mice were intragastrically treated with NEPP for 8 days before APAP(265mg·kg-1)injection except for normal group.Serum levels of alanine ALT,AST and the liver of biochemical indicators and hepatic histopathology was detected routinely.Method Selectin of serum in TNF-α,IL-6,IL-1β expression level was detected by ELISA method.Results:1)The levels of ALT,AST in blood and MDA in hepatic tissue has been decreased obviously in NEPP groups(P<0.05),but significantly increase the levels of SOD,and histological changes of liver were observed.2)NEPP could significantly decrease the levels of AST and ALT in CCl4-induced liver injured mice,reduce the levels of MDA,TGF-β1,TNF-α,IL-6,IL-1β content,IL-1β,JNK,P38-MAPK,TNF-αgene and protein expression levels significantly decreased.Significantly increase the levels of SOD,GSH,GSH-PX and histological changes of liver were observed.3)As compared with the model control group,BEVD medium dose group could significantly decrease the levels of AST and ALT in APAP induced liver injured mice and decrease TNF-α,IL-6,and IL-1 p,reduce the levels of MDA content,but significantly increase the levels of SOD,GSH-PX,and GSH in serum and liver tissues.Conclusion:1)NEPP has the function of protecting liver cells.2)NEPP can obviously decrease liver injury,which may be related with decreased levels of inflammatory factors induced by CC14,increased the anti-oxidant ability,and reduced oxidative stress,and NEPP can induce changes of MAPK signaling in and inhibin gene expression in liver injury mice.3)NEPP can obviously decrease liver injury,which may be related with decreased levels of inflammatory factors induced by APAP,increased the anti-oxidant ability,and reduced oxidative stress. |
