Different Prescriptions Of Jiawei Chai Shaoyao Liujun Tang Comparative Study On The Effect Of Liver Depression And Spleen Deficiency Model IBS-D Rat

Objective: To observe the effect of "Jiawei Chai Shaoyao Liujun decoction" on visceral sensitivity and serum serotonin 5-hydroxytryptamine(5-HT)and substance P(SP)in rats with liver depression and spleen deficiency IBS-D model by gavage and enema,to compare the efficacy of Jiawei Chai Shaoyao Liujun decoction and to explore the mechanism of its treatment for liver stagnation and spleen deficiency IBS-D.Methods: 40 healthy SD rats were randomly divided into normal group,experimental group(including model group,desut group,chinese medicine gavage group,chinese medicine enema group).In addition to the normal group,the model was constructed by the method of "Senna leaf irrigation combined with chronic restraint stress ".After successful modeling,the rats in each group except normal group were treated with drug intervention for 2 weeks.;after intervention treatment,rectal balloon dilatation was selected to measure the minimum volume threshold of rat AWR(abdominal wall retreat reflex)in each group to evaluate rat visceral sensitivity.;After anesthesia,rat serum was collected and detected by ELISA.5-HT?SP content in mouse serum.;After the experiment,the rats were put to death by dislocation,dissecting and separating the colon at the end of the rat,cutting about 0.5 cm of the colon terminal tissue at the same site,and making pathological sections to observe the morphologicalchanges of the colon mucosa in the rats.Results:(1)Compared with the normal group,the rats were tired of mental burnout,decreased activity,decreased food intake and drinking water,increased defecation frequency,short interval of defecation,dark brown water stool or paste stool,no mucus,blood stool,dirty bedding,obvious anal fecal dye,stool pinch.The body weight loss,visceral sensitivity and serum SP?5-HT content of the model rats were significantly increased(P<0.05);(2)The general conditions of Deshute group,Chinese medicine enema group and Chinese medicine gavage group were all improved in different degrees after treatment,and the body mass growth of Chinese medicine gavage group and Chinese medicine gavage group was significantly higher than that of model group(p<0.05);(3)After treatment,the visceral sensitivity and 5-HT?SP content of rats were significantly lower than those of model group(P<0.05);(4)Chinese medicine enema group had a more significant effect on reducing visceral sensitivity and lowering 5-HT?SP in rat serum than those in Desut group and Chinese medicine gavage group(P<0.05).conclusion :(1)A IBS-D rat model with liver depression and spleen deficiency syndrome was successfully established by "Senna leaf irrigation and stomach combined with chronic restraint stress ".The model has low cost,easy operation,high stability and strong replicability;(2)" Jiawei Chai Shaoyao Liujun decoction" can reduce the content of 5-HT?SP substance in serum of rats with liver depression and spleen deficiency IBS-D improve diarrhea,increase the body weight of rats,increase the minimum threshold of AWRvolume,reduce the high sensitivity of viscera and achieve the therapeutic effect on IBS-D rats with liver depression and spleen deficiency.Treatment IBS-D effective drug delivery methods,clinical drug use to provide a choice of drug delivery and approach;(3)this experiment speculated that "Jiawei Chai Shaoyao Liujun decoction" treatment of liver stagnation and spleen deficiency type IBS-D the possible mechanism is to reduce the content of 5-HT?SP in model rat serum,regulate intestinal fluid,electrolyte and water secretion,inhibit intestinal spasm and contraction,reduce intestinal visceral perception,thereby improving the symptoms of abdominal pain,diarrhea,achieve IBS-D treatment.Among them," Jiawei Chai Shaoyao Liujun Tang " enema group has the best effect,may be with the intestinal administration,drug direct to the hospital,in the local retained a high concentration,while avoiding the destruction of drug activity and the first-pass effect of the liver,improving the bioavailability of the drug;(4)the results of this experiment showed that "Jiawei Chai Shaoyao Liujun decoction" gavage,enema treatment of liver depression and spleen deficiency type IBS-D rats have obvious curative effect.Confirmed that "liver stagnation spleen deficiency dampness Sheng" is the etiology and pathogenesis of liver stagnation spleen deficiency type IBS-D,"soothing liver and invigorating spleen,winning dampness and stopping diarrhea,relieving pain " is its treatment principle,in line with clinical practice.
