?Aims?By comparing the ischemic stroke patients' transcriptome sequencing in acute and recovery phase analysis result,the possible mechanism of the recovery of neurological impairment in ischemic stroke was explored.?Method?This study adopt the longitudinal study.6 ischemic stroke patients was conducted,who met the criteria of sodium exclusion.Venous blood was collected from these patients including the acute and convalescent stages.Total RNA of the whole blood was extracted for transcriptome sequencing,and analyzed differential genes expression by using GO analysis and KEGG metabolic pathway analysis.?Result?1.There were a total of 290 differential expressed genes in the convalescent and acute transcriptome sequencing 177 expressed genes increased and 113 expressed genes decrease.2.Through using GO analysis,differential expressed genes were mainly enriched in intracellular structure composition,including mitochondrial respiratory chain complex,histone deacetylation regulation,nuclear transcription mRNA catabolism process,T cell differentiation regulation,innate immune activation signal transduction and so on.3.Through using KEGG metabolic pathway analysis,the raised gene mainly enriched in toll-like receptor signal transduction pathways,IL-17 signal transduction pathway,NOD signaling pathways,NF Kappa signaling pathway and MAPK signaling pathways,EB virus infection,nerve nutriment pathways and vascular endothelial growth factor signal pathway,lysosome,sheath lipid signaling pathways,vascular endothelial growth factor signaling pathway and so on.The decreased gene mainly enriched in ribosome,systemic lupus erythematosus(sle),alcohol poisoning,cancer-causing virus,DNA replication and protein output,huntington's disease,oxidative phosphorylation,Parkinson's disease,base excision repair,glycine,serine and threonine metabolism,alzheimer's disease,homologous recombination,pyrimidine metabolism,nucleotide excision repair,nonalcoholic fatty liver disease and so on.?Conclusion?The recovery of neurological function in ischemic stroke was mainly related to inflammation,mitochondrial respiratory chain composition,cell division and proliferation,nerve regeneration and other pathways.
