[Objective]By using lectin microarray technique,we analyzed the difference of serum glucose and egg sugar chain spectrum between patients with stage I,II,III and IV of lung cancer and normal volunteers,in order to find out the structural markers of sugar chain related to the occurrence and development of lung cancer.It is helpful for early diagnosis and evaluation of lung cancer.[Methods]Firstly,the glycoprotein expression profiles of serum samples from 54 patients with different stages of lung cancer(early stage I and II 9,III 24,IV 21)and 48 healthy volunteers were analyzed by lectin microarray technique.HCA cluster analysis and Ratio value analysis were used to compare the glycoproteins in serum of lung cancer patients and healthy volunteers,and lectin imprinting technique was used to reverse verify the results of lectin microarray.Then the ROC curve was used to evaluate the potential value of differential lectin-recognized sugar chain structure in the early diagnosis of lung cancer.[Results]The relative abundance of lectin ACA,GSL-I,NPA,PSA,PTL-II,SJA,STLand VVA in serum of lung cancer patients was significantly higher than that of healthy volunteers(foldchange > 1.5,p < 0.05).The relative abundance of the glycan structure recognized by lectin EEL and RCA120 in patients with lung cancer was significantly lower than that in healthy volunteers(foldchange < 0.67,p < 0.05).2.There were significant differences in the glycan structure recognized by lectin PWM and EEL in stage III and between IV lung cancer patients and healthy volunteers(p < 0.001).The results of lectin imprinting of EEL showed that The abundance of lectin EEL recognized Gal ? 1-3(Fuca 1-2)Gal(blood group B antigen)in normal volunteers and patients with early lung cancer(especially 70 k Da and 50 k Da)was higher than that in III and IV patients.EEL's ROC curve analysis showed that lectin E The structure of the sugar chain identified by EL can effectively distinguish between the early stage and the stage III(AUC = 0.917,p<0.001),and the patients with lung cancer(AUC = 0.926,p <0.001).3.The glycan structure recognized by lectin GSL-I,STL was only significantly different between patients with early lung cancer and healthy volunteers(P < 0.05).The results of ROC curve analysis of lectin GSL-I showed that the glycan structure recognized by lectin GSL-I was used to distinguish early lung cancer from normal volunteers.When AUC=0.723,p=0.034,suggested ? Gal NAc,? Gal,recognized by lectin GSL-I in serum,it was suggested that lectin GSL-I recognized ?Gal NAc,? Gal,in serum.The structure of anti-A and B sugar chain can effectively distinguish early lung cancer patients from healthy volunteers.4.The relative abundance of man ? 1-6Man structure recognized by lectin NPA in all lung cancer patients was significantly higher than that in normal volunteers(foldchange > 1.5,p < 0.05).The results of lectin imprinting of NPA showed that the abundance of the glycan structure recognized by lectin NPA in serum proteins(especially 65 k Da and 55 k Da)of all patients with lung cancer was higher than that of healthy volunteers.The results of ROC curve analysis of NPA showed that the glycan structure recognized by NPA was used to distinguish AUC=0.816,p between patients with early lung cancer and healthy volunteers.5.The relative abundance of ?-Gal,Gal ?-1,4Glc NAc structure recognized by RCA120 in all patients with lung cancer was significantly lower than that in normal volunteers,and the ROC curve showed that AUC=0.802,p=0.004;in distinguishing early stage lung cancer patients from normal healthy volunteers was found.It is suggested that the ?-Gal,Gal ?-1,4Glc NAc sugar chain structure recognized by lectin RCA120 in serum can effectively distinguish early lung cancer patients from healthy volunteers.[Conclusions]1.The corresponding glycan structure recognized by lectinACA,PSA,PTL-II,SJA,STL,VVA and EEL may be helpful in the diagnosis of lung cancer.2.Among them,?-Gal,Gal ?-1 recognized by lectin RCA120,4Glc NAc sugar chain structure,Mana 1-6Man sugar structure recognized by NPA,? Gal NAc,? Gal,anti-A and B sugar structure recognized by GSL-I may be helpful to the early diagnosis of lung cancer.Gal ?1-3(Fuca 1-2)Gal(blood group B antigen)recognized by lectin EEL may be useful in differential diagnosis of early and middle-advanced lung cancer.
