Objective: To investigate the effect of knocked down expression of KAI1 gene on the biological function of lung adenocarcinoma cells and the possible molecular mechanism involved.Methods: The expression of KAI1 gene in KAI1 blank group and negative control group was detected by real-time fluorescence quantitative PCR and protein imprinting.The expression of KAI1 gene in KAI1 blank group and negative control group was detected by real-time fluorescence quantitative PCR and protein imprinting;MTT kit method and Transwell chamber invasion and migration technique were used to detect the changes of proliferation,invasion and migration ability of lung adenocarcinoma cells after the knocked down expression of KAI1 gene;Real-time fluorescence quantitative PCR and Western blot were used to detect the changes of mRNA and protein expression levels of KAI1,Axin1 and GSK3? in each group after the knocked down expression of KAI1 gene;The expression of KAI1,Axin1 and GSK3? in human-related cancer tissues was detected by using the Elivison TMplus method;The lentivirus-mediated KAI1 overexpression gene was injected into nude mice to observe the effect of lentivirus-mediated overexpression gene on the growth of human lung adeno-carcinoma cell line A549.The effect of lentivirus on the growth of tumor in nude mice was observed,ten female nude mice were randomly divided into two groups: blank plasmid group and experimental group.Observe and record the living condition of naked mice,the growth of transplanted tumors,and measure the length and short diameter of two groups of transplanted tumors every 3 days.The average volume of the tumor was calculated and the corresponding growth curve was drawn.After 5 weeks of continuous observation,the nude mice were killed and the tumor tissue was stripped off.The weight of the tumor tissue was measured,the volume of the tumor was measured.Results: The mRNA expression level and protein expression level of KAI1 gene in KAI1 expression group was lower than that in blank group and negative control group,and the difference was statistically significant(p<0.05),The results of MTT assay showed that the cell proliferation ability of the KAI1 decreased expression group was significantly higher than that of the blank group and the negative control group,and the difference was statistically significant(p <0.05),The results of Transwell chamber invasion test showed that the number of cells passing through Matrigel matrix glue in KAI1 knocked down expression group was significantly higher than that in blank group and negative control group,and the difference was statistically significant(p<0.05),The results of real-time fluorescence quantitative PCR and Western blot showed that the expression of Axin1 and GSK3? in KAI1 gene expression group were lower than that in blank group and negative control group,but the expression level of vimentin and Slug were higher in KAI1 gene expression group than that in blank group and negative control group.The growth curve of transplanted tumor in nude mice showed that the growth rate of transplanted tumor was faster than that of experimental group(p<0.05).Conclusions: The knocked down expression of KAI1 gene can significantly promote the proliferation,invasion and migration of lung cancer cells.The overexpression of KAI1 gene can effectively inhibit the growth of human lung adenocarcinoma cells A549 and H1299 in nude mice;the knocked down expression of KAI1 gene can inhibit the expression of Axin1 and promote the expression of vimentin.KAI1 gene may inhibit the invasion and migration of human lung adenocarcinoma cells by regulating the Wnt signaling pathway.
