Objective.To investigate the regulation and mechanism of HS3ST2 gene on the expression of Wnt pathway and ECM-receptor interaction pathway related molecules in rat GH3 cell line.Methods.The expression of HS3ST2 gene in rat GH3 cells was interfered.The m RNA transcription level and protein expression of Wnt pathway and ECM receptor interaction pathway related genes(?-catenin,WIF1,LEF1,SDC4)were detected by Western-Blot and quantitative real-time PCR(RT-q PCR).Results.After interfering with the expression of HS3ST2 gene in GH3 cells,the m RNA and protein expression level of HS3ST2 gene decreased,and the expression level of its modified heparan sulfate proteoglycan protein decreased.The m RNA transcription and protein expression level of ?-catenin and WIF1 genes in Wnt signaling pathway decreased(P<0.05),while the m RNA and protein expression level of WIF1 gene increased(P<0.05).The m RNA and protein expression of SDC4 gene in ECM receptor interaction pathway increased(P<0.05).Conclusion.HS3ST2 gene promotes the proliferation and invasiveness of GH3 cells by regulating the expression of related genes of Wnt signaling pathway and ECM receptor interaction pathway.It provides experimental basis for finding new therapeutic targets and preventive drugs for GH pituitary adenoma.
