Purpose:In this study,ornithine decarboxylase(ODC)and antizyme inhibitors(AZI)were determined in cumulus cells at oocyte retrieval day to explore the relationship between ODC、AZI expression and oocyte quality and its corresponding embryo development potential,so as to establish a new method for predicting oocyte quality.Methods:This is a retrospective study of 112 infertility patients who received assisted reproductive technology in the Reproductive medicine center of the Jiangsu province people’s hospital from June 2019 to January 2020.All subjects included in the study have signed informed consent.We collected cumulus cells at oocyte retrieval day,and used western blot method for determination of ODC and AZI relative expression level in cumulus cells.(1)firstly,We analyzed the correlation between ODC and data related to the basic clinical conditions of the patients and IVF-related data of patients;Then,the quartile method was used to divide patients into four groups:Q1,Q2,Q3,Q4,and compare the differences between the four groups in data related to the basic clinical conditions of the patients and IVF-related data.(2)In the same way,We analyzed the correlation between AZI and data related to the basic clinical conditions of the patients and IVF-related data of patients;Then,the quartile method was used to divide patients into four groups: Q1,Q2,Q3,Q4,and compare the differences between the four groups in data related to the basic clinical conditions of the patients and IVF-related data.(3)In the end,we analyzed the correlation between cumulus cells ODC and AZI.Results:(1)ODC and AZI existed in person cumulus cells,and there were differences in expression between individuals.In this study,We detected ODC expression in 53 people,AZI expression in 76 people,ODC and AZI expression in 17 people.(2)There was no correlation between ODC and data related to the basic clinical conditions of the patients.(3)ODC was positively correlated with the number of high-quality embryos(r=0.294,P=0.034)and the rate of high-quality embryos(r=0.346,P=0.012),and had no correlation with the remaining IVF related data.(4)The quartile method was used to divide ODC into Q1(<0.689),Q2(0.689-1.570),Q3(1.570-2.346),Q4(2.346≤).There were differences in the infertility years(P=0.027)and basis testosterone(P=0.028)among the four groups.There was no difference in the remaining clinical data.The rate of egg maturation(P<0.001)and the rate of normal fertilization(P=0.008)was lower in group Q1 and higher in group Q2,Q3,Q4.The rate of high-quality embryos(P=0.006)was lower in group Q1 and Q2,and higher in group Q3 and Q4.The remaining IVF-related data showed no difference between the four groups.(5)There was no correlation between AZI and data related to the basic clinical conditions of the patients.(6)AZI was positively correlated with the number of high-quality embryos(r=0.297,P=0.010)and the rate of high-quality embryos(r=0.336,P=0.003),and had no correlation with the remaining IVF related data.(7)The quartile method was used to divide AZI into four groups: Q1(<0.726)Q2(0.726-1.11)Q3(1.11-1.765)Q4(1.765≤).BMI(P=0.036)and Gn amount(P=0.025)were different among the four groups,and there was no difference in the remaining data related to the basic clinical conditions of the patients.The number of high-quality embryos(P=0.033),the cleavage rate(P=0.007),the rate of transplanting embryos(P=0.003)and the rate of high-quality embryos(P<0.001)were gradually increased in the four groups of Q1,Q2,Q3,Q4,and the difference was statistically significant.(8)ODC was positively correlated with AZI,and the difference was statistically significant(r=0.552,P=0.032).Conclusions:(1)ODC and AZI existed in person cumulus cells,and there were differences in expression between individuals.(2)The cumulus cells ODC was positively correlated with the number of high-quality embryos and the rate of high-quality embryos,indicating that granulosa cell ODC was of certain value in predicting oocyte quality and its corresponding embryo development potential.(3)The cumulus cells AZI was positively correlated with the number of high-quality embryos and the rate of high-quality embryos,suggesting that AZI was a good indicator for evaluating oocyte quality and its corresponding embryo development potential.(4)The cumulus cells l ODC was positively correlated with AZI,and the determination of cumulus cells ODC and AZI could enhance the value of predicting oocyte quality and its corresponding embryo development potential.
