| Gastric cancer is the most common malignant gastrointestinal tumor which originates in the gastric mucosal epithelium.Recent studies have found that polyunsaturated fatty acid(PUFA)has very significant effects on inhibiting or alleviating the occurrence and development of various chronic diseases. In this topic, natural products ω-6 fatty acid with with high biologically activity arachidonic acid(AA),linoleic acid(LA),ω-3 fatty acid docosahexenoic acid(DHA),eicosapentaenoic acid(EPA),normal gastric cell GES-1,gastric cancer cells MGC-803 and SGC-7901 were performed for incubation. And their effects on proliferation, invasion, migration and apoptosis of gastric cancer cells were investigated. The results showed that the inhibiting effects of PUFA(LA,AA,DHA and EPA)on gastric cancer cells GES-1,gastric cancer cells SGC-7901 and MGC-803 showed concentration dependence.160 μmol/L and 200 μmol/L PUFA(LA,AA and EPA)could significantly inhibit the activity of normal gastric cells and gastric cancer cells. There was almost no effect of DHA with different concentrations(40 to 200 μmol/L)on the activity of normal gastric cells. But it could inhibit the growth of gastric cancer cells SGC-7901 and MGC803 to some extent.LA,AA and EPA could significantly promote the apoptosis of normal gastric cells GES-1(P<0.05).There was no obvious promoting apoptosis effect of DHA on normal gastric cells GES-1(P>0.05).LA,AA,EPA and DHA could promote apoptosis of gastric cancer cells SGC-7901 and MGC-803 to some extent(P<0.05).DHA and EPA could inhibit the invasiveness and migration of gastric cancer cells SGC-7901 and MGC-803 to some extent(P<0.05).The test results showed that there were obvious side effects of LA,AA and EPA on normal gastric cells when inhibiting malignant biological behavior of gastric cancer cells SGC-7901 and MGC-803 at the same time. There was no significant effect of DHA on activity of normal gastric cells, which was expected to be an anticancer drug.It provided theoretical basis for the treatment of clinical gastric cancer. |
