| Objective:Preterm birth(PTB)is defined as a term of birth between 28 weeks and less than 37 weeks of gestation.About a third of PTB premature rupture of membranes is incomplete preterm(preterm prematurely ruptured membranes,PPROM).Up to now,the pathogenesis of PPROM has not been clarified completely.The differential expression of long chain non-coding RNAs(lncRNAs)in placental cells of PTBs and PPROM was explored to understand their possible involvement in the pathogenesis of PPROM.A new possibility is provided to study the pathogenesis of PPROM.Methods:A total of 40 cases of maternal placenta samples aged from 25 to 30 years old were collected.Group B,FTB group,39-40 weeks gestation,no membrane rupture;No membrane rupture was observed in group C and PTB at 35 weeks gestation.Group D,PROM(premature rupture of membrane)39-40 weeks gestation.RNA was extracted from the placental tissue of the pregnant women,and the different expression of LncRNAs and mRNAs was screened by LncRNA microarray detection.The differentially expressed genes were analyzed by GO(Series Test of Cluster)and Pathway analysis and subsequent STC(Series Test of Cluster)analysis.Results:LncRNAs were detected in the placental microarray of PPROM(group A)in 1954,776 and 1050,which were significantly different from those in full term birth(group B),PTB(group C)and PROM(group D),respectively.The data of the four experimental groups were analyzed and compared,including the regulation and down-regulation of pathway A and B,A and C,and A and D.Our results showed that the expression of 22 pathways was up-regulated by 7,up-regulated by 18 and down-regulated by 15 in A and C,respectively.The expression of A was up-regulated by 33 and down-regulated by 7 in A and b.Functional analysis showed that inflammatory response,ECM(Extracellular matrixc)-receptor interaction,apoptosis,actin cytoskeleton and smooth muscle contraction were the main pathogenesis of PPROM.Conclusion:The identification of different expression of LncRNAs in human placenta of FTB,PTB,PPROM and PROM indicates that lncRNAs may be involved in the pathogenic progression of human pregnancy related to reproductive status and reproductive disorder.Further studies on the metabolic pathways revealed that infection and inflammation,ECM-receptor interaction,apoptosis and smooth muscle contraction were the main pathogenic mechanisms involved in the development of PPROM,PROM and PTB. |
