| [Objective]To investigate the expression of ENO1 in colorectal cancer tissues and its correlation with clinicopathological features of colorectal cancer patients.By knocking down the expression of ENO1 in colorectal cancer cells by lentivirus,the effect of targeted regulation of ENO1 on the proliferation and metastasis of colorectal cancer cells was observed.[Methods]The expression of ENO1 in 113 colorectal cancer tissues and 32 adj acent normal tissues normal tissues was detected by Immunohistochemistry(IHC).The expression level of ENO1 in eight colorectal cancer cells and the influence of ENO1 knockdown on EMT in colorectal cancer cells were detected by Western blot.Lentivirus transfection technology was used to establish cell line in which ENO1 was stably downregulated.Colony formation assay and cell migration assay were used to evaluate the effects of ENO1 knockdown on the proliferation and metastasis of colorectal cancer cells.[Results]ENO1 expression in human colorectal cancer tissues was upregulated,and its high level of expression was associated with tumor size,lymph node metastasis and TNM stages.Colorectal cancer cells expressed higher ENO1 in protein level.Lentivirus can effectively knock down the expression level of ENO1 in colorectal cancer cells;Silencing ENO1 inhibited the proliferation and migration of colorectal cells;Silencing ENO1 increased E-cadherin expression but decreased Vimentin expression.[Conclusions]ENO1 expression in human colorectal cancer tissues was significantly higher than that in adjacent tissues,and was associated with tumor size,differentiation,lymph node metastasis,and TMN stages.Cell in vitro experiments suggested that ENO1 is involved in regulating colorectal cancer cell proliferation and metastasis.The effect of ENO1 on colorectal cancer cell metastasis may be modulating the EMT progress. |
