Objectives:By exploring the relationship between the expression of non-SMC condensin ? complex subunit G(NCAPG)gene in epithelial ovarian cancer tissues and ovarian cancer cells,the prognosis of patients,clinicopathological characteristics and ovarian cancer cell proliferation ability,the biological function and clinical significance of NCAPG in the malignant phenotype of ovarian cancer cells were clarified Significance,provide theoretical basis and experimental basis for NCAPG gene as a potential new target for ovarian cancer diagnosis,prognosis monitoring and intervention.Methods:1.Retrieve NCAPG related information based on Oncomine database,analysis of the expression of NCAPG gene in ovarian cancer,Kaplan-Meier Plotter database was used to analyze the survival of patients,and to explore its relationship with the prognosis of patients with ovarian cancer.2.Immunohistochemistry(IHC)was used to detect the expression level of NCAPG protein in 70 cases of ovarian cancer tissues and 10 cases of normal tissues in epithelial ovarian cancer tissue microarray chips,and analyzed the correlation of NCAPG expression with age.International Federation of Gynecology and Obstetrics(FIGO)stage,tissue differentiation and lymphatic metastasis in ovarian cancer patients.3.Real-time Quantitative polymerase chain reaction(RT-qPCR)was used to detect the expression of NCAPG mRNA in human ovarian epithelial cells(IOSE80)and human ovarian cancer cells(A2780,SKOV3 and OVCAR-3).4.Culture human ovarian cancer OVCAR-3 and SKOV3 cells,divided into shRNA-NCAPG group and shRNA-NC group,transfected with shRNA-NCAPG and shRNA-NC.The CCK-8 method was used to detect the proliferative capacity of two groups of cells,and to explore the effect of NCAPG gene on the proliferative capacity of cells.Results:1.There are 10 studies on NCAPG gene and ovarian cancer in the Oncomine database,Meta-analysis found that the mRNA level of ovarian cancer NCAPG was significantly higher than that of normal ovarian tissue(P<0.01);Kaplan-Meier Plotter survival analysis found that patients with high expression of NCAPG gene had significantly lower Progression-free Survival(PFS)and Overall Survival(OS)than those with low expression(P<0.01)2.The results of IHC detection of ovarian cancer microarray tissue chip showed that the positive expression rate of NCAPG in 70 cases of epithelial ovarian cancer tissue was 62.86%,and the positive expression rate in 10 cases of adjacent ovarian tissue was 20.00%,Expression rate is higher than the adjacent tissues(P<0.05);The expression level of NCAPG is significantly higher in patients with ovarian serous carcinoma,late FIGO stage,poorly differentiated,and lymphatic metastatic ovarian cancer tissues(P<0.05).3.Using qRT-PCR to detect NCAPG mRNA levels in human ovarian cancer cells and normal human ovarian cells,the results showed that the expression of NCAPG in human ovarian cancer cells OVCAR-3 and SKOV3 cells was significantly higher than that in human normal ovarian IOSE80 cells(P<0.001);Compared with IOSE80 cells,A2780 cells had no statistically significant difference in NCAPG mRNA expression(P>0.05)4.shRNA-NCAPG and shRNA-NC were transfected into ovarian cancer OVCAR-3 and SKOV3 cells,and the CCK-8 method was used to detect the proliferation ability of OVCAR-3 and SKOV3 cells(P<0.01).Conclusions:The expression of NCAPG gene is significantly increased in epithelial ovarian cancer tissues and ovarian cancer cells,and its high expression is closely related to histological type,tumor progression,poor patient prognosis,tumor cell proliferation,and plays the role of oncogene in ovarian malignant tumors.It is expected to become a potential new target for ovarian cancer diagnosis,prognosis monitoring,and intervention,which deserves further study.
