Perilla(Perilla frutescens(L).britt)is a traditional multi-purpose cash crop in China and one of the first batch of 60 Chinese medicines promulgated by the Ministry of Health as both food and medicine.It contains a variety of physiologically active chemical ingredients.In this paper,the extraction process,content analysis and hypoglycemic properties of flavonoids in perilla were studied.The article takes perilla as the research object,the main research content includes:the microwave-assisted extraction method is used to perform a single factor test on the perilla root,stem and leaf,and the optimal extraction process parameters are selected through the optimization of response surface analysis.Secondly,the ultraviolet spectrophotometer method was used to analyze the dynamic study of the flavonoid content of perilla in different provenances during the growth period,and to explore the correlation between the content and the climatic factors in Harbin.Finally,perilla flavonoids are devoted to development and utilization to explore their hypoglycemic properties.The specific test results are as follows:Taking the extraction rate of flavonoids as the evaluation index,the single-factor test was used to determine the optimal extraction process conditions of flavonoids from the microwave-assisted extraction method.It is finally concluded that the optimal extraction conditions for the extraction of perilla flavonoids by microwave-assisted method are 70-90%ethanol volume fraction,the liquid-to-material ratio is 10:1-20:1 mL/g,the microwave time is 25-35min,and the microwave power 120-385W.The microwave extraction process of flavonoids was improved by using response surface Box-Benhnken optimization technology.The best technological conditions were as follows:the best technological parameters of perilla root were microwave time 30.2min,microwave power 253.22W,liquid-to-material ratio 13.40:1mL/g,flavonoid extraction rate 4.41%;The best process parameters are microwave time 30.81min,microwave power 272.84W,liquid-to-material ratio 16.31:1mL/g,flavonoid extraction rate 2.27%;the best process parameters of perilla leaves are microwave time 30.16min,microwave The power is 269.49W,the liquid-to-material ratio is 14.97:1mL/g,and the flavonoid extraction rate is 7.83%.The flavonoid content of perilla from different provenances was analyzed by ultraviolet spectrophotometer.It was finally concluded that as far as perilla roots are concerned,the highest total flavonoid content in roots of various origins occurred in mid-July,and the total flavonoid content of Yunnan provenances was higher than that of other provenances;for perilla stems,The highest total flavonoid content in stems of various sources was produced at the end of June,and the total flavonoid content in Gansu provenances was significantly higher than that of other provenances;as far as perilla leaves were concerned,the highest total flavonoid content in leaves of various sources In mid-July,the total flavonoid content in Gansu provenances was significantly higher than in other provenances.By analyzing the correlation between the content of perilla flavonoids and local climatic factors,it is found that the content of flavonoids has a certain correlation with the changes of local climatic factors.Among them,temperature,sunshine time and rainfall have significant effects on it.Sunshine time is positively correlated with it,and rainfall is negatively correlated with flavonoid content.This paper also studied the hypoglycemic effect of flavonoids extracts from roots,stems and leaves of Perilla frutescens on normal mice,and the effect of different doses of extracts from roots,stems and leaves on blood glucose of mice.The results showed that the extracts from the roots and stems of Perilla frutescens had little effect on the blood glucose of mice.The flavonoid extracts from the leaves of Perilla frutescens could significantly reduce the blood glucose of normal mice at a dose of 600mg/kg.bw,which had a certain auxiliary effect on blood glucose.The blood sugar of mice decreased with the dose of 400mg/kg.bw,but the effect was much smaller than that of the high dose group.When the flavonoid extract of perilla leaf was 200mg/kg.bw the blood glucose of mice did not change obviously because of the low dosage.
