Objective:We investigated the expression of coenzyme A:cholesterol acyltransferase 1(ACAT1)in normal gastric cells and four types of gastric cancer cells,as well as the expression of ACAT1 in gastric cancer and adjacent cancer tissues,to preliminarily explore the correlation between the expression of AC AT1 and the clinicopatho logical characteristics of gastric cancer patients,providing a new idea and method for the diagnosis and treatment of gastric cancer.MethodsWestern Blot was performed to investigate the expression of ACAT1 in one normal gastric cell(Ges-1)and four gastric cancer cells(AGS,SNU-1,MKN45,HGC-27);and then Immunohistochemistry was performed to investigate the expression of AC AT1 in 82 cases of gastric cancer tissue and adjacent tissues who had not received radiotherapy or chemotherapy before surgery were collected,and all of them were diagnosed as gastric cancer by pathology department after surgery.Differences of ACAT1 expression in gastric cancer tissue and adjacent tissues were analyzed,and investigate the correlation between the expression of AC AT1 and the clinicopathological characteristics of gastric cancer patients.Results1.The results of Western Blot showed that the expression of ACAT1 protein in four gastric cancer cells was significantly higher than that in normal gastric cells(P<0.05).2.The results by Immunohistochemistry method showed that positive expression rate of gastric cancer tissues was significantly higher than that of adjacent cancer tissue,74.39%and 20.73%,and the difference was statistically significant(P<0.05).3.Chi-square analysis showed that the expression level of AC AT1 protein was associated with TNM-stage(?2=6.448,P=0.011),lymph node metastasis(?2=9.464,P=0.002).There is no relationship with gender,age,tumor diameter,differentiated degree,tumor location,M-stage.ConclusionThe expression of ACAT1 protein in four gastric cancer cells was higher than that in normal gastric tissue cells.The expression of ACAT1 in gastric cancer tissues was higher than that in adjacent cancer tissue.The expression level of ACAT1 was correlated with the TNM-stage and lymph node metastasis,but not with the patient's gender,age,tumor diameter,differentiated degree,tumor location and M stage.
