Background:Endometrial carcinoma is a common female reproductive system disease.In recent years,the morbidity and mortality are increasing year by year and showing a younger trend.Although remarkable progress has been made in the early detection and treatment of endometrial cancer,it can still be detected that a large number of patients with advanced diseases have a poor prognosis,the lack of effective drug treatment and the metastasis of endometrial cancer limit the treatment of endometrial cancer.Therefore,it is imperative to find a wide range of applicable,safe and effective anti-tumor drugs.As a kind of phytoestrogen,kaempferol belongs to flavonoids.Compared with other flavonoids,kaempferol has different biological characteristics,especially in anti-tumor.Kaempferol has inhibitory effects on a variety of estrogen-related malignant tumors.However,the effect of kaempferol on endometrial carcinoma is not clear.Purpose:The main purpose of this study was to investigate the selective anticancer effects of Kaempferol against MFE-280 endometrial carcinoma cells along with evaluating its effects on apoptotic pathway,cell cycle phase distribution,cell invasion,cell migration and PI3K/Akt/mTOR signalling pathway.Methords:Cell viability of MFE-280 cells was assessed by MTS assay.Apoptosis was determined by AO/EB double staining.Cell cycle distribution was determined by flow cytometry.Boyden chamber assays were performed to study the effects of Kaempferol on cell migration and invasion.Western blotting was used to investigate the expression of PI3K/Akt/ m-TOR pathway.Results:Compared with control treatment,Kaempferol treatment significantly inhibited the viability(P<0.05),and promoted apoptosis(P<0.05)in MFE-280 cells.Kaempferol treatment significantly increased the G2/M phase cells percentage(P<0.05).The cell migration and invasion were dramatically suppressed in cells treated with Kaempferol when compared with that of control treatment(P<0.05).Kaempferol treatment significantly downregulated the phosphorylation of PI3 K,AKT and mTOR(P<0.05).Conclusion:Kaempferol palys an important role in the viability,apoptosis,cell cycle distribution,cell migration and invasion.The mechanism may be related to the supression of PI3K/AKT/mTOR activity.
