Part ?: Increase of NK cells and abnormal phenotype in peripheral blood of patients with knee osteoarthritisObjective To investigate the distribution and phenotypic characteristics of natural killer(NK)cells in peripheral blood of knee osteoarthritis(KOA).Methods Flow cytometry was used to detect and analyze the distribution of lymphocyte subsets in the peripheral blood of healthy people without KOA and KOA patients,and to compare the distribution of NK cells of different.phenotypes and their surface function receptors,human leukocyte antigen DR(HLA-DR).,Natural cytotoxic receptor 3(NCR3 / NKP30),CD69 and NKG2 D expression.Results The proportion of CD3-CD56 + NK cells in peripheral blood of KOA patients was significantly up-regulated(P <0.01),and CD3-CD56 + CD16 + NK cells increased mainly(P <0.01).The expressions of CD3 + CD4 + T cells and CD3 + CD19 + B cells were down-regulated(P <0.05),while CD3 + CD8 + T cells decreased slightly without significant difference(P> 0.05).The expression of HLA-DR molecules on the surface of NK cells in KOA was up-regulated,while the expression of NKP30 was significantly reduced.Conclusion The proportion of NK cells in peripheral blood of KOA patients increased,and the expression of CD16 and HLA-DR was up-regulated and the expression of NKP30 was down-regulated.Part ?: Expression of very late antigens in peripheral blood and synovial tissue of knee osteoarthritis.Objective: To determine the expression status of very advanced families in peripheral blood and mononuclear cells of synovial tissue in patients with osteoarthritis of the knee,in order to improve our understanding of the possible mechanisms of the inflammatory process.Methods: Peripheral blood from patients with knee osteoarthritis and non-osteoarthritis control group who met the inclusion criteria and synovial tissue from patients with joint replacement were isolated and mononuclear cells were labeled with fluorescent antibodies and detected by flow cytometry Expression of integrin ?1?1 to ?6?1(VLA-1 to 6).Results: The expression of VLA-3 in peripheral blood NK of KOA was significantly higher than that in the control group.The heavy NK cells in synovial tissues were a large number of CD56 brights,which were significantly higher than those in peripheral blood.There are more VLA-1,VLA-3,and VLA-6 expression on the surface of synovial tissue NK cells.Conclusion: The aggregation of natural killer cells in synovial tissue may be related to the VLA family.
