Objective:To investigate the expression levels of serum and decid ual histidine-rich glycoprotein(HRG),decidual expression of proge sterone receptors and in different age groups(older pregnancy group,young pregnancy group)of mice during second pregnancy decidua cell morphological changes,by comparing different ages,using low mole cular weight heparin sodium(LMWH)or not,HRG expression levels in decidua and serum,and decidual expression levels of proges terone receptors,further clarify expression differences in treatment groups and control groups of pregnant mice of different ages,whe ther there is a correlation between the expression level of HRG in decidua and serum,and the correlation between the effect of LMWH and HRG on decidualization.Further to demonstrate the difference in age expression of HRG and its effect on decidualization.Methods:We randomly divided 8-10-week-old(young group)/ 8-10-month-old(older group)ICR female mice into four groups: young treatm ent group(n = 10)/young control group(n = 10);Older treatment group(n = 15)/older control group(n = 15).Female rats were separated from 8-10-week-old male mice when they had a vaginal plug after being in the same cage.It was regarded as the 0.5th day of pregnancy,then the teatm ent groups/the control groups were treated with LMWH(Enoxaparin)or PBS on the 4.5th day.The pregnant mice were sacrificed by cervical dislocation after blood was drawn from the posterior orbit,and the uterus of the pregnant mices were removed on the 14.5th day.The ELISA was used to measure the HRG levels in the four groups of serum,and the immunohistochemical method was used to detect the HRG expression level and PR level in the decidua and at the same time observe thedecidual morphology.Statistical analysis was performed to explore whether low-molecular-weight heparin sodium could improve the quality of decidua by interfering with the coagulation mechanism of HRG in decidua.Rusults:(1)Comparison of serum HRG expression levels: The average expression levels of HRG in the serum of the teatment group of the elderly pregnancy group were higher than those of the control group,and the difference was statistically significant(P<0.05).The average expression level of HRG in the serum of the young treatment group was higher than that of the control group,and the difference was not statistically significant(P>0.05).Compared with teatment groups of different ages and control groups,the serum expression level of HRG incr eased with age,and the difference was statistically significant(P <0.05)..(2)There was a negative correlation between HRG expression in serum and HRG level in decidua between the treatment groups /the control groups in the elderly group(r =-0.88 /-0.86,P<0.05).There was no significant correlation between HRG expression in serum and HRG in decidua between the treatment groups and the control groups in the young group(r =-0.40 /-0.26,P> 0.05).(3)The average positive expression level of PR in the treatment group was higher than that in the control group,and the difference was not statistically significant(P> 0.05);Between the treatment groups /the control groups,the average positive expression level of PR in decidua of the young groups were higher than that in the older groups,and the differences were statistically significant(P < 0.05).(4)Compared with the decidual morphology of the treatment groups and the control groups in different age groups,the decidua deve loped better in the treatment groups.Conclusion:(1)The Low-molecular-weight heparin sodium can improve the quality of decidua by interfering with the coagulation and antifibrinolysis of HRG in decidua,and the effect is stronger with age.Serum HRG expression gradually increased with age.Decr eased serum HRG levels at the same age may be related to poor decid ualization.(2)Declining PR expression in decidua with age may be related to poor decidualization.Although low-molecular-weight heparin does not increase the expression level of decidua PR,it can improve the qua lity of decidua by affecting the morphology of decidua.
