Aspongopus chinensis Dallas,1851 is a special medical insect belonging to Dinidoridae,Hemiptera,Insecta.It was firstly reported in the Compendium of Materia Medica and used to treat bronchitis,stomach pain,sexual dysfunction,endometriosis,and so on.Modern pharmacology proved its pharmacological effects of anti-cancers,antibacterial,and so on.Hemolymph from A.chinensis could significantly inhibit the proliferation of human breast cancer cells and gastric cancer cells in vitro.However,animal model test,as an important midway to a potential new medicine,is not carried out by now.In this study,based on its inhibitory effect on mouse breast cancer 4T1 cells in vitro,animal model was established by injecting mouse breast cancer 4T1 cells,and the anti-cancer effects of A.chinensis hemolymph alone and in combination with cisplatin were analysised.The research methods and results were as follows:1.Effect of hemolymph from A.chinensis on the growth of mouse breast cancer4T1 cells in vitro4T1 cells of mouse breast cancer were treated with hemolymph from A.chinensis.Morphological observation showed that the hemolymph of A.chinensis could promote their shrinkage and inhibit their proliferation;MTT assays showed that the hemolymph could significant inhibit the proliferation of breast cancer cells in a timeand concentration-dependent manner;Hoechst 33258 staining test showed that the proportion of apoptotic cells increased with the increase of the time;flow cytometry test showed that with the increase of the concentration of hemolymph,the proportion of cells in S phase increased significantly,and the proportion of cells in G1 phase decreased significantly,which suggested that hemolymph may block breast cancer cells in S stage.2.Effect of A.chinensis hemolymph by intragastric administration on the growth of mouse breast cancer xenografts in vivoMouse model of breast cancer transplanted tumor were built successfully,and the effects of different concentrations of A.chinensis hemolymphon by intragastric administration on the breast cancer xenograft were explored by the analysis of the changes in the mouse weight,the volume breast cancer tumors,the weight of breast cancer tumors,liver,spleen,lung,and kidney,the number of blood cells and the structure of tissues.The results showed that intragastric administration was not effective in inhibiting the size and weight of breast cancer xenografts,and had no significant effect on the body weight,liver,spleen,lung,and kidney.It did not significantly enhance the inhibiting effect of cisplatin on breast cancer xenografts,and also did not alleviate its side effects on mouse body.3.Effect of A.chinensis hemolymph by intraperitoneal injection on the growth of mouse breast cancer xenografts in vivoThe same methods as showed in 2 were performed to carry out the same contents.Meanwhile,Hoechst 33258 staining was used to observe the apoptosis of tumor tissue.The results showed that intraperitoneal injection of hemolymph can significantly inhibit the growth of breast cancer xenografts,and induce apoptosis of tumor tissues.The side effects of cisplatinon on the reduction of body weight,liver,spleen and thrombocytopenia in mice were alleviated by the hemolymph,but it could not enhance the inhibitory effect of cisplatin on breast cancer.Conclusions:The hemolymph of A.chinensis could inhibit breast cancers effectively in vivo and in vitro,and without significant side effects,at the same time,it could alleviate the side effects of cisplatinon.
