| Objective: To observe the changes of glycolipid metabolism in the high-fat induced obese male mice model.To investigate the relationship between Kisspeptin/GPR54 and abnormal glycolipid metabolism caused by high fat-induced obesity through detecting the levels of leptin receptor,Kisspeptin/GPR54 protein in pancreas,liver and adipose tissue of high-fat induced obese male mice.Methods: Forty-eight 4/5-week-old C57BL/6J male mice were selected as the research objects,and they were randomly divided into a control group of 12(fed basic feed)and a high-fat group of 36(fed high-fat feed).After 19 weeks of feeding,the mice in the high-fat group were divided into diet induced obesity(DIO)and diet induced obesity-resistance(DIO-R)groups.The 12 of the top heaviest in the 36 mice fed high-diet were defined as DIO Group,and the 12 lightest were defined as DIO-R Group.All mice were sacrificed after taking blood from the inferior vena cava,and the absolute weights of liver,kidney,perirenal fat,and peritesticular adipose tissue were weighed and the relative weights were calculated;HE staining was used to observe the pathological changes of liver,fat,and pancreas tissue;Blood glucose levels in mouse serum were measured by Glucose Meter;Serum triglycerides(TG),total cholesterol(TC),high-density lipoprotein cholesterol(HDL-C),low-density lipoprotein cholesterol(LDL-C),and liver Triglyceride level(TG)were measured by biochemical method;Serum insulin and leptin levels and calculate insulin resistance index were detected by ELISA method;Kisspeptin,GPR54,Ob-R protein levels in pancreatic tissue were immunohistochemically detected;Total protein from liver tissue and adipose tissue were extracted and the levels of Kisspeptin,GPR54,Ob-R protein in liver and adipose tissue were detected by Western blot.Results:1.After 19 weeks of experiment,the weight of the mice in the DIO group was significantly higher than that in the control group and the DIO-R group(P<0.05).The weight of the mice in the DIO-R group was not significantly different from that in the control group(P>0.05);2.The food intake by mice in the high-fat diet group was higher than that of the normal diet group,and the water consumption was lower than that of the ordinary diet group(P<0.05);3.The absolute weight of liver and kidney of the DIO group mice was significantly higher than that of the control group,and the liver and kidney coefficients were lower than those of the control group(both P<0.05).Compared with the control group,liver weight of DIO-R group was found no different from that of the control Group(P>0.05);the liver coefficient was lower than that of the control group,while the kidney weight and kidney coefficient of the DIO-R group were higher than those of the control group(all P<0.05).Compared with the DIO-R group,the DIO group had no significant differences in perinephric fat,perirenal fat coefficient,testicular fat,and testicular fat coefficient(all P>0.05);4.HE staining revealed that steatosis was seen in the liver tissue of DIO group and DIO-R group mice.Compared with the control group,the fat cell cytoplasm increased in the DIO group.There were no pathological changes in HE staining of the pancreas of the three groups of mice;5.TG,TC and LDL-C in the serum of the DIO group and DIO-R mice were higher than those in the control group(all P<0.05).There was no difference in HDL-C levels of the mice in the group(P>0.05);6.The blood glucose levels of the DIO and DIO-R mice were significantly higher than those of the control group(P<0.05),the insulin levels of the DIO group were higher than those of the control group(P<0.05),and there was no difference between the insulin levels of the DIO-R and control groups(P>0.05),the insulin index of the DIO group was higher than that of the control group(P<0.05),while there was no difference in the insulin index of the DIO-R group and the control group(P>0.05);7.The serum leptin levels in the DIO and DIO-R groups were significantly higher than those in the control group(P<0.05);8.The triglyceride levels in the liver of DIO group mice were significantly higher than those in the control group(P < 0.05).There was no significant difference in the triglyceride levels of the DIO-R group and the control group(P>0.05);9.The expression levels of Kisspeptin and Ob-R proteins in the pancreas of DIO and DIO-R mice were significantly lower than those in the control group(both P < 0.05).There was no significant difference of GPR54 protein expression levels of the mice in the three groups(P>0.05);10.The expression levels of Kisspeptin,GPR54 and Ob-R proteins in the liver of DIO and DIO-R mice were significantly higher than those in the control group(all P<0.05);11.The expression levels of Kisspeptin,GPR54 and Ob-R proteins in the adipose tissue of DIO and DIO-R mice were significantly lower than those in the control group(all P<0.05);Conclusion: There were glycolipid abnormality in high-fat-induced obesity male mice.The expression levels of Kisspeptin/GPR54 and leptin receptor protein in liver,fat and islet of obese mice have changed,which may be related to the abnormal glucose and lipid metabolism caused by high-fat induced obesity. |
