Objective: To study the expression of different transcripts of DDB2 gene in gastric cancer and distant normal gastric tissues and their effects on the biological behavior of gastric cancer cells.Methods: Sixty-eight pairs of gastric cancer tissues and their corresponding distant normal gastric tissues were selected to make tissue chips.The expression differences of WT-encoded proteins in normal transcripts of gastric cancer and normal tissues were analyzed by immunohistochemical staining.Then,total RNA of 48 pairs of gastric cancer and distant normal gastric tissues was extracted and qPCR was used to detect the expression of WT and D1 transcripts of DDB2 gene.the expression of different transcripts of DDB2 in various gastric cancer cell lines was detected by agarose gel electrophoresis and qPCR.DDB2 WT and D1 overexpression plasmids were constructed,and the empty vector and the two overexpression plasmids were transfected into gastric cancer cell lines.WB were used to detect the transfection.BGC823 and AGS cell lines were selected as tool cell lines based on the transfection and its basic expression.The cell proliferation and migration ability of each group was detected by CCK-8 and transwell.Epithelial-mesenchymal transition and apoptosis were found by WB experiments.Results: There was no significant difference of DDB2 WT transcript-encoding protein expression between gastric cancer and normal tissues.At the RNA level,the expression of DDB2 D1 transcripts in gastric cancer tissues was significantly higher than that in distant normal tissues(P <0.01),while WT transcripts showed no significant difference between the two.In vitro experiments,WT transcripts promoted the proliferation of gastric cancer cells but inhibited their migration(P <0.05),while D1 transcripts suppressed proliferation but promoted their ability of migration(P <0.05).Both transcripts had no significant effect on the apoptosis and Epithelialmesenchymal transition of the cells.Conclusion: DDB2 D1 transcripts are enhanced in gastric cancer tissues.WT transcripts promote proliferation and inhibit migration,while D1 transcripts inhibit proliferation and promote migration.
