| Objective:With a high morbidity and mortality,gastric cancer is one of the most common malignant tumors worldwide.Therefore,it is particularly important to explore new biomarkers of gastric cancer for the early diagnosis and accurate targeted therapy of gastric cancer.In addition,since the tumor immunotherapy has shown promising prospect in recent years,exploring the relationship between tumors and immunity and its molecular mechanism can bring new hope for tumor patients.CMTM6(CKLF like MARVEL transmembrane domain containing 6),which was thought to be related to sperm evolution and circadian rhythm system,belongs to the chemokine-like factor gene superfamily(CKLFSF).It was reported that CMTM6 can stabilize PD-L1 expression on the membrane,but the exact relationship between CMTM6 and tumor immunity remains to be studied.In addition,the prognostic value of CMTM6 is still controversial.This study aimed to explore the expression of CMTM6 in gastric cancer,its correlation with PD-L1 and other tumor immune-related factors,and whether it can be used as a biomarker for diagnosis and prognosis.Materials and methods:In this study,information from different databases(Oncomine database,Kaplan Meier Plotter database,TISIDB database,GEPIA database)was used to analyze the expression level of CMTM6 in gastric cancer,its relationship with tumor immune-related cells and molecules,and its relationship with the prognosis of gastric cancer patients.The expression of CMTM6 in gastric cancer patients was also analyzed by immunohistochemical staining in tissue microarray(TMA)including 806 pairs of gastric cancer tissues.The correlation between CMTM6 and clinicopathological data and prognosis was analyzed,and its correlation with PD-L1 was verified in TMA.We also explore its potential prognostic value in gastric cancerResults:We found that although the differential expression of CMTM6 was not obvious in the Oncomine database,the expression level of CMTM6 in gastric cancer tissues was significantly lower than normal tissues in our Tissue Microarray.Besides,the higher expression of CMTM6 indicated a better prognosis in the Kaplan Meier Plotter database In addition,our analysis found and verified that CMTM6 is not only significantly related to PD-L1,but also significantly related to a variety of tumor immune cells and molecules In addition,CMTM6 can be used as a satisfying diagnostic marker for gastric cancer,and we found that the combination of CMTM6 and PD-L1 has better diagnostic efficacy. |
