Objective: Neurotrophin 3(NTF3)is involved in numerous biological processes,especially the survival and differentiation of neuronal cells.However,its research in solid tumors is not much and its role in hepatocellular carcinoma(HCC)is not well examined.With the continuous updating and improvement of biological information technology,this study obtained the expression level and survival prognosis of NTF3 in liver cancer by big data analysis.And by set different expression levels of NTF3 to conduct in vitro experiments to investigated the NTF3 function in HCC progression and revealed the underlying molecular mechanisms.Methods: We use The Cancer Genome Atlas(TCGA)database bioinformatical analysis of publicly available data.To explore the correlation between the expression level and prognositive relevance of NTF3 in hepatocellular carcinoma and its adjacent tissues.Take 30 pair clinical specimens and use real-time quantitative PCR(Quantitative real time polymerase chain reaction,qRT-PCR)to verify the expression level of NTF3 in hepatocellular carcinoma,and at the same time,Immunohistochemistry of paraffin specimens of HCC tissues were performed for exploring the expression of NTF3.Cell growth and proliferation were analyzed by Cell Counting Kit-8(CCK-8)assay.Cell invasion and migration were analyzed by Boyden transwell and wound healing assays.Protein expression and mRNA levels(n=80)were evaluated by Western Blot and qRT-PCR.Cell apoptosis was evaluated by flow cytometry.Results: The TCGA database showed that NTF3 expression was significantlly lower in HCC tissues compared to adjacent non-tumors liver tissues,and the difference was statistically significant(P <0.001).Lower NTF3 expression was significantly associated with decreased patient overall survival.The results of qRT-PCR and IHC showed that the expression of NTF3 in HCC tissues was overall lower.NTF3 overexpression reduces the proliferation,migration and invasion of HCC cell lines.NTF3 overexpression reduced the proliferation,migration,and invasion of HCC cell lines.And lower NTF3 expression was significantly associated with clinicopathological features.Conclusions: According to TCGA database analysis and clinical sample verification,decreased expression of NTF3 is involved in poor prognosis of patients with HCC by promoting HCC cell proliferation,migration,and invasion.Our findings provided novel understanding into the pathogenesis of HCC and the role of NTF3 in tumor progression,suggesting that targeting NTF3 has potential therapeutic and diagnostic value for HCC treatment.And provided an experimental basis for future research.
