Objective: The chemokine receptor CXCR5,as a member of the CXCR subfamily of chemokines,is not participated in B cell maturation and lymphocyte homing,but also plays an irreplaceable role in tumor’s progression and prognosis of patients.In this paper,we studied the expression of CXCR5 in renal cell carcinoma and analyzed the correlation between that and pathological parameter,to reveal the role of CXCR5 in the occurrence and development of renal cell carcinoma,also it can provide a therapeutic targets and prognostic markers for the renal cell carcinoma.Methods:This study collected the clinical data of 55 patients who underwent radical nephrectomy or nephron sparing surgery from May 2015 to may 2018 in the urology center of Qingdao Municipal Hospital,and collected the cancer tissue and the paracancerous tissues(2 cm away from cancer lesion),then detected the expression of CXCR5 protein in the renal cell carcinoma tissue and normal tissue near the cancer by immunohistochemistry,Data processing through SPSS21 software package,further analysis the relationship between the expression of the CXCR5 and the clinicopathological features of renal cell carcinoma,such as gender,age,tumor size,lymph node metastasis,TNM stage and Fuhrman grade.To evaluate the role of CXCR5 in the occurrence and progression of renal cell carcinoma.Results: 1.The expression of CXCR5 protein was mainly located in the cytoplasm and nuclear membrane of cancer cells;the positive expression rate of CXCR5 protein was 69.1%in 38 of 55 cases of renal cell carcinoma,and it was basically strong positive;while only 3 of the 20 cases showed positive expression in para-cancer normal tissues,and the positive expression rate was 15%,which was basically weak positive.Compared with the normal tissues,the positive expression rate in renal cell carcinoma was significantly higher,the difference was statistically significant(P = 0.000 < 0.01).2.The expression of CXCR5 was related to tumor TNM stage and Fuhrman grade.The positive expression rate of CXCR5 in advanced RCC(TNM Ⅲ-Ⅳ stage)was higher than that in localized RCC(TNM Ⅰ-Ⅱ stage),the difference was statistically significant(P =0.029 < 0.05).The positive expression rate of CXCR5 in middle and low-differentiated RCC(Fuhrman grade Ⅲ-Ⅳ)was higher than that in well-differentiated RCC(Fuhrman grade I-II),the difference was statistically significant(P = 0.011 < 0.05).3.There was no correlation between the expression of CXCR5 and patients’ gender,age,tumor size,lymph node metastasis(P > 0.05).Conclusion:The expression of CXCR5 protein in renal cell carcinoma tissue was significantly higher than that in normal tissues,and it is correlated with TNM stage and Fuhrman grade,but is not correlated with gender,age,tumor size and lymph node metastasis.This suggests that CXCR5 might be correlated with the occurrence,development and metastasis of renal cell carcinoma,and it can be used as a biomarker for the prognosis evaluation of RCC,and provide a potential target for the diagnosis and treatment of RCC.
