With wide applications of phellinus linteus in medicine,the identification of the growth years of phellinus linteus has become particularly important.In this thesis,a scanning electron microscope(SEM)was used to observe and measure the fruit body microstructures of phellinus linteus artificially-cultivated for 1-3 years old.The microstructures of mycelia and spores were observed on the micro-nano scale,and the morphologic characteristics of mycelia and spores in different years were obtained.The varying patterns of mycelia and spores of phellinus linteus in different growth years were summarized through measurements.A classifier was trained to classify the images by year based on the previous obtained microstructural images of the context and tubes of phellinus linteus.Before the classifier was trained,the image problems such as burrs,bright and dark fringes were preprocessed.Then,the texture feature extraction methods based on the gray level co-occurrence matrix,SIFT feature and the bag-of-words(BOW)model were used to extract the image features,and the support vector machine(SVM)was used as a classifier for training.The features extracted by two methods and the new features combined based on the former two features to train the SVM classifier were used,and the three classifiers were obtained.Finally,the weighted voting was conducted on the three classifiers to obtain the final result.The final classification was tested,and the accuracy rate of classification of combined features directly using SVM classifier was 77.778 %,while the accuracy rate of classification based on the integrated learning classifier was 88.889 %.Through the experimental results,the combined learning classifier had a better classification effect with the accuracy of88.889 %.The fruit body extract of phellinus linteus of different growth years was added to esophageal carcinoma cells,and the cells were studied by scanning electron microscope(SEM)and atomic force microscope(AFM),respectively.The microscopic images of the living cells and the solidified cells were obtained and the mechanical properties of the living cells were analyzed.The extract of which growth year of phellinus linteus being more effective on esophageal cancer cells was determined.Through the comparison and analysis,it was found that the liquid extract of phellinus linteus in one year had more obvious effect on esophageal cancer cells than that in two or three years.With the combined application of multiple microscopes,multiple information of cancer cells can be obtained more accurately,which provides a new way for future medical detection,which is of great importance for applications.
