With the rapid development of social economy and the changes of people’s living habits,diseases are becoming more and more diversified.Currently,the death rate of cardiovascular is increasing each year,and cardiovascular disease is the biggest killer threatening human health,which mainly because of hypertension.Synthetic drugs are the most drugs used to treat hypertension on the market.Long-term usage of synthetic drugs has great negative effects on human health,therefore it is especially important to develop a drug or health supplement which is safe without side effects.Kelp is a common natural marine plant with many kinds of activities.Although Chinese kelp products rank first in the world these years,the help is not fully utilized leading to the waste of resources.Medicinal bacteria have multiple medicinal functions and various biological activities.For instance,they have a biological fermentation function,which can ferment the substrate containing nutrients into a growth substrate and transfer into active substance with an increased biological activity.In this study,the fermented product was obtained by fermenting kelp with medicinal bacteria,and the antihypertensive activity of fermented product was discovered.On the one hand,it developed an innovative way in drug developments in blood pressure reduction and health care.On the other hand,the study provided the ideas for deep utilization and development of both marine resources and medicinal fungi resources.In this article,antihypertensive activity was used as a measurable indicator to screen suitable high-efficiency strains and extraction methods by different levels of hypotensive activity of kelp products.Then different fermented products were obtained by fermenting kelp with high-efficiency strains in different days,and the fermentation time was selected by fermentation yield,extraction rate and antihypertensive activity.Finally,fermentation products were extracted and purified to study the blood pressure lowering activity of different components,and many substances were examined.Firstly,the angiotensin-converting enzyme inhibition model in vitro was used to screened out 3-3T and 6-2Y samples having high blood pressure lowering activity with different strains and different extraction methods.The 3-3T sample was obtained only from fermented kelp with strain No.3 by the 3T method,and 6-2Y was obtained only from the fermented kelp with strain No.6 by the 2Y method.Secondly,the kelp was fermented with strains No.3 and No.6,respectively,and the samples were collected every 5 days after the strain was overgrown the bottle.The strain No.6 was sampled four times,and the strain No.3 was sampledeight times.After two batches of fermentation and activity detection,the fermentation yield,the extraction rate and blood pressure lowering activity were used as indicators to screen the optimal fermentation time.The optimal fermentation time of the No.3 strain and strain No.6 were 35 d and 15 d,respectively.Finally,the fermentation products fermented by the No.3 strain were extracted,and the obtained extract products were separated into individual components according to the molecular weight level by ultrafiltration,and then the blood pressure lowering activity of each component was determined to determine the main distribution range of active substance,which indicated that the active substances are mainly distributed in less than 3k Da components.The content of polysaccharides,proteins,polyphenols and sulfates in each component were determined,and the relationship between the content of each component and the blood pressure lowering activity was studied and analyzed,which provides a theoretical and data basis for future separation and identification of components.Through the research on the blood pressure lowering activity of the fermented kelp products,the fermentation process and the extraction conditions,it is found that the blood pressure lowering activity of kelp can be significantly improved by fermentation,and the utilization rate of kelp can also be increased,indicating that the products obtained from the fermented kelp by medicinal bacteria have high medicinal value and practical value.It is a green and environmentally friendly way to develop and utilize marine resources,which has high research value in the pharmaceutical and environmental fields.
