| Objective: To explore the therapeutic effect of aucubin on type 2 diabetic mice through NF-κB signaling pathway.Method: C57BL/6J mice were fed with common feed for 7 days and started to be modeled.All mice were divided into 5 groups on average: blank control group,type 2diabetes model group,and low dose of peach glucoside(1 mg/kg)The treatment group,the middle-dose arbutin(5 mg/kg)treatment group and the high-dose arbutin(10 mg/kg)treatment group.The blank control group was fed with common mouse feed,and the remaining groups were fed with high-fat feed for 28 days.After 28 days of feeding,streptozotocin was intraperitoneally injected for modeling.The mice with successful modeling started the treatment of aucubin: different doses of aucubin were intraperitoneally injected once a day for 28 consecutive days.After the treatment,all the experimental mice were tested for glucose tolerance and insulin tolerance;they were sacrificed,and the serum was taken to detect the four items of serum lipid,insulin and inflammatory factors,and the insulin resistance index and insulin sensitivity index were calculated;the pancreas HE staining was used to observe the shape of the islet;Western blotting to detect the protein content of P-IκB,NF-κBp65 and p-NF-κBp65.Results: After treatment,the body weight of the pyophyllin low,medium and high dose treatment group was significantly higher than that of the type 2 diabetes model group(p<0.001),respectively increased by 9.4%,9.8%,12.7%,and 10 mg/kg peach leaf The coat color,urine output and mental state of the mice in the coral glycoside treatment group were close to those in the normal group;at the same time,the blood glucose and the type 2 diabetes model group of the peach leaf glycoside low,medium and high dose groups were significantly reduced(p<0.001),decreased by 26.3%,42.5%,and 54.2%,respectively;TG of mice with low,medium,and high doses of aucubin significantly decreased(p<0.05),and TC,LDL-C,and HDL-C decreased The trend was not statistically significant;HOMA-IR of mice in the low-,medium-,and high-dose aucubin groups was significantly reduced(p<0.05),with a decrease of 42.86%,55.66%,and 66.24%,respectively.ISI was significantly increased(p<0.05);the levels of serum insulin,TNF-α,IL-6 and IL-1β in mice with low,medium and high doses of aucubin were significantly lower than those in type 2 diabetes model group,But there was no significant difference;NF-κB pathway-related proteins were significantly reduced in mice with low,medium,and high doses of pyophyllin(p<0.05);compared with mice in the diabetes model group,pyophyllin The structure and morphology of the islets of mice in the group were improved.Conclusion: Pyophyllin can regulate the disorder of body weight,blood sugar and blood lipids in mice with type 2 diabetes and reduce insulin resistance.Pyophyllin can regulate the serum insulin and inflammatory factor levels in mice with type 2 diabetes.In addition,the level of NF-κB signaling pathway-related proteins decreased after peach glucosinolate treatment,indicating that peach glucosinolate has a therapeutic effect on mice with type 2diabetes. |
